液质联用研究微生物肥料对脐橙果实品质的影响

摘  要:为明确微生物肥料对脐橙果实品质的影响,分别施用微生物肥料(microbial fertilizer,MF)和化肥(chemical fertilizer,CF)的纽荷尔脐橙果实为研究对象,测定其主要理化指标,并采用超高效液相色谱串联四级杆飞行时间质谱(UPLC-Q/TOF-MS)研究了汁胞中的代谢物。结果表明:MF组果实的重量、体积、果皮色泽指数,汁胞的可滴定酸、可溶性糖、维生素C、总酚及总黄酮含量均显著高于CF组;UPLC-Q/TOF-MS从汁胞中共鉴定出10类79种代谢物,利用正交偏最小二乘判别分析和单因素方差分析,共37种显著差异代谢物被鉴定出,其中MF组相较于CF组有17种物质含量显著增加,黄酮类物质占比最大,其次是有机酸类物质;利用KEGG通路富集分析共注释到6条代谢途径,包括类黄酮生物合成途径、柠檬酸循环、丙酮酸代谢途径、半乳糖代谢途径、C5-支链二元酸代谢途径和酪氨酸代谢途径。研究结果表明,MF可通过影响相关代谢途径调节果实中有机酸、糖类、酚类、黄酮类等代谢物含量,从而提高脐橙果实品质。To verify the effects of microbial fertilizer(MF)on the quality of navel orange fruits,this study focuses on the Newhall navel orange fruits treated with MF and chemical fertilizer(CF),respectively.Firstly,the main physical and chemical indicators of navel orange fruits were determined.Then,the metabolites in juice sacs were detected based on ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(UPLC-Q/TOF-MS).Thefindings demonstrated that the weight,volume and color index of fruit peel,and titratable acid,soluble sugar,vitamin C,total phenolic and totalflavonoid contents of juice sacs were significantly higher in MF than CF.A total of 79 metabolites,belonging to 10 categories,were identified based on UPLC-Q/TOF-MS.A total of 37 differential metabolites were screened out by orthogonal partial least squares-discriminant analysis and analysis of variance.Among them,17 substances in MF significantly increased compared with CF,andflavonoids accounted for the largest proportion,followed by organic acids.The differential metabolites were primarily enriched in 6 metabolic pathways,includingflavonoid biosynthesis,citrate cycle,pyruvate metabolism,galactose metabolism,C5-Branched dibasic acid metabolism and tyrosine metabolism,according to the KEGG enrichment analysis.The results showed that MF could regulate the content of organic acids,sugars,phenolics,flavonoids and other metabolites in fruit by influencing related metabolic pathways,so as to improve the quality of navel orange fruit.
