摘 要:目的研究右美托咪定在新生大鼠海马区域依托咪酯麻醉条件下对PI3K/Akt信号传导途径的影响。方法选取雄性SD大鼠80只,依据随机数字表法随机分为8组,每组10只,按照不同处理方式分别记作N组(生理盐水)、I组(脂肪乳剂)、D组(二甲基亚砜)、E组(依托咪脂60 mg/kg)、DE-1组(右美托咪定25μg/kg+依托咪脂60 mg/kg)、DE-2组(右美托咪定50μg/kg+依托咪脂60 mg/kg)、DE-3组(右美托咪定75μg/kg+依托咪脂60 mg/kg)、LY294002组(LY294002+右美托咪定75μg/kg+依托咪脂60 mg/kg)。于大鼠苏醒2 h后,每组取5只用透射电镜观察海马神经元的形态学变化,剩余5只采用Western blot法检测各组海马Akt和pAkt(ser473)蛋白含量。结果8组大鼠Akt蛋白含量差异无统计学意义(P>0.05)。相较于E组、I组、D组,DE-1组、DE-2组、DE-3组和LY294002组pAkt(ser473)蛋白含量更低(P<0.05);与E组比较,DE-1组、DE-2组、DE-3组和LY294002组pAkt(ser473)蛋白含量更高于E组(P<0.05);与DE-3组比较,DE-1组、DE-2组、LY294002组pAkt(ser473)蛋白含量更低于DE-3组(P<0.05);LY294002组pAkt(ser473)蛋白含量高于DE-1组(P<0.05)。大鼠海马神经元细胞结构损伤随着右美托咪定剂量的增加而减轻。结论右美托咪定可能通过减轻对PI3K/Akt信号通路的抑制作用,来缓解依托咪酯对发育期大鼠海马神经元造成的损伤。Objective Study on the effect of dexmedetomidine on PI3K/Akt signaling pathway under etomidate anesthesia in the hippocampus of neonatal rats.Methods According to the random number table method,80 male SD rats were selected and randomly divided into 8 groups,10 rats in each group,denoted as Group N(saline group),Group I(fat emulsion group),Group D(dimethyl sulfoxide),Group E(etomidate 60 mg/kg),Group DE-1(dexmedetomidine 25μg/kg+etomidate 60 mg/kg),Group DE-2(dexmedetomidine 50μg/kg+etomidate 60 mg/kg),DE-3 group(Dexmedetomidine 75μg/kg+Etomidate 60 mg/kg),LY294002 group(LY294002+Dexmedetomidine 75μg/kg+Etomidate 60 mg/kg)according to different processing methods After the rats were awakened for 2 h,5 rats in each group were taken to observe the morphological changes of hippocampal neurons by transmission electron microscopy,and the remaining 5 rats were tested for hippocampal Akt and pAkt(ser473)protein content by western blot method.Results There was no statistically significant difference in Akt protein content among the 8 groups(P>0.05).pAkt(ser473)protein content was significantly lower in groups E,DE-1,DE-2,DE-3 and LY294002 than in groups N,I and D(P<0.05);pAkt(ser473)protein The content of pAkt(ser473)protein was significantly higher than that in group E(P<0.05);the content of pAkt(ser473)protein was significantly lower in group DE-1,DE-2 and LY294002 than that in group DE-3(P<0.05);the content of pAkt(ser473)protein in group LY294002 was significantly higher than that in group DE-1(P<0.05).The cytoarchitectural damage of rat hippocampal neurons was reduced with the increase of dexmedetomidine dose.Conclusions Dexmedetomidine may alleviate the damage caused by etomidate to hippocampal neurons in developing rats by reducing its inhibitory effect on the PI3K/Akt signaling pathway.
