摘 要:目的 探讨电针对对氯苯丙氨酸(4-chloro-DL-phenylalanine, PCPA)致失眠大鼠乳脂球表皮生长因子Ⅷ(Milk fat globule-epidermal growth factor Ⅷ,MFG-E8)的影响。方法 50只SD大鼠(雌雄各半)随机抽取10只作为空白组,除空白组外,剩余40只均腹腔注射PCPA建立失眠模型,经造模、筛选随机分为模型组、电针组、艾司唑仑组,每组10只,观察大鼠一般情况变化,戊巴比妥钠睡眠试验观察大鼠睡眠潜伏期和睡眠持续时间,经电针、艾司唑仑药物干预7d后,酶联免疫法(ELISA)检测大鼠血清中MFG-E8含量,Western blot法检测下丘脑组织中MFG-E8蛋白表达。结果 经PCPA造模干预的失眠模型大鼠,戊巴比妥钠睡眠实验显示,睡眠潜伏期延长(P<0.01),睡眠持续时间缩短(P<0.05);与空白组比,模型组大鼠血清MFG-E8水平降低(P<0.05),下丘脑中MFG-E8蛋白相对表达量下降(P<0.01);经电针、艾司唑仑药物干预后,失眠大鼠昼夜节律明显改善,体质量明显升高(P<0.05),血清中MFG-E8水平升高(P<0.01);下丘脑中MFG-E8蛋白相对表达增高(P<0.01);电针组与艾司唑仑组之间MFG-E8含量无明显统计学差异(P>0.05)。结论 电针对PCPA致失眠大鼠睡眠障碍具有改善作用,揭示电针可上调MFG-E8的表达,其可能机制与MFG-E8调控小胶质细胞,逆转M1表型极化,降低致炎因子水平有关。Objective:To study the effect of electroacupuncture on milk fat globule-epidermal growth factor VIII(MFG-E8)in rats with insomnia induced by 4-chloro-DL-phenylalanine(PCPA).Methods:Fifty SD rats(equally divided by gender)were randomly selected,with 10 rats serving as the blank group.The remaining 40 rats were administered PCPA via intraperitoneal injection to establish an insomnia model.After modeling,the rats were randomly divided into the model group,electroacupuncture group,and estazolam group,10 rats each group.The general conditions of the rats were observed,and the pentobarbital sodium sleep test was used to measure sleep latency and sleep duration.After 7 days of intervention with electroacupuncture or estazolam,MFG-E8 levels in the serum of the rats were measured by using ELISA,and the relative expression of MFG-E8 protein in the hypothalamus was detected by Western blot.Results:In the PCPA-induced insomnia model rats,the pentobarbital sodium sleep test showed prolonged sleep latency(P<0.01)and shortened sleep duration(P<0.05).Compared with that of the blank group,the serum MFG-E8 levels in the model group were reduced(P<0.05),and the relative expression of MFG-E8 protein in the hypothalamus decreased(P<0.01).After intervention with electroacupuncture and estazolam,the circadian rhythm of insomnia rats significantly improved,and body weight increased(P<0.05).MFG-E8 levels in the serum increased(P<0.01),and the relative expression of MFG-E8 protein in the hypothalamus also increased(P<0.01).No significant statistical difference in MFG-E8 levels was observed between the electroacupuncture and estazolam groups(P>0.05).Conclusion:Electroacupuncture improves sleep disorders in PCPA-induced insomnia rats,suggesting that electroacupuncture may upregulate the expression of MFG-E8.The underlying mechanism might be related to MFG-E8's regulation of microglial cells,reversal of M1 phenotype polarization and reduction of pro-inflammatory cytokine levels.
