摘  要:目的探讨甘草对心脏衰老的作用以及其发挥作用的潜在靶点。方法通过挖掘GEO数据库,获取小鼠心室RNA测序基因集;通过Herb网站挖掘甘草相关靶基因,DAVID网站进行差异基因通路富集分析,Metascape网站进行差异基因功能富集分析。通过60μM H_(2)O_(2)诱导心肌细胞衰老,细胞衰老β-gal染色评估衰老细胞数量;通过qRT-PCR检测衰老标志物p21表达、甘草靶基因与心脏衰老差异基因共有基因表达。结果在GEO数据库中挖掘到基因集GSE12480,其与甘草靶基因有4个共有基因,这些基因在心肌细胞衰老模型中表达上调;甘草提取物可以减少60μM H_(2)O_(2)诱导的心肌细胞中衰老细胞数量,降低心肌细胞p21表达,并降低VCAM1、ICAM1和PTGS1的mRNA表达。结论甘草提取物可以减轻H_(2)O_(2)诱导的小鼠心肌细胞衰老,减少衰老细胞数量,降低p21表达,且其发挥作用可能与其降低VCAM1、ICAM1和PTGS1的表达有关。Objective To investigate the effect of licorice root on heart aging and the potential targets for its action.Methods By mining the GEO database,the gene sets of ventricular RNA sequencing in young and old mice were obtained.The licorice-related target genes were mined through the Herb website,and a differential gene pathway enrichment analysis was performed through the DAVID website while a differential gene function enrichment analysis was conducted through the Metascape website.Cardiomyocyte aging was induced by 60μM H_(2)O_(2),and the number of aging cells was assessed byβ-gal staining.The expression of aging marker p21 and the expression of shared genes of licorice target genes and heart aging differential genes were detected by qRT-PCR.Results The gene set GSE12480 was found in the GEO database,which shared four common genes with licorice target genes,and these genes were up-regulated in expression in the cardiomyocyte aging model.Licorice extract reduced the number of aging cells and decreased the expression of cardiomyocytes p21 in cardiomyocytes induced by 60μM H_(2)O_(2),decreased the mRNA expression of VCAM1,ICAM1,and PTGS1.Conclusion Licorice extract attenuates H_(2)O_(2)-induced aging in mice cardiomyocytes,reduces the number of aging cells,and decreases p21 expression,and its action may be related to its reduction in the expression of VCAM1,ICAM1,and PTGS1.
