摘 要:骨髓间充质干细胞(bone marrow stem cells,BMSCs)离体培养可诱导分化为软骨细胞、成骨细胞、脂肪细胞等,广泛参与骨再生和修复,维持骨代谢平衡。BMSCs成骨分化是一个涉及多系统、多基因、多信号通路的复杂生理过程,包括Notch、ERK、Hedgehog等信号通路。本文综述这些信号通路在BMSCs成骨分化过程中各自的组成及传导机制,以期为骨质疏松症及骨缺损等疾病的基因辅助治疗筛选关键性的基因靶点,并探索新的治疗思路。Bone marrow stem cells(BMSCs)are able to differentiate into various cell types in vitro like chondrocytes,osteoblasts and adipocytes,which are widely involved in bone regeneration and repair,and maintain the balance of bone metabolism.The osteogenic differentiation of BMSCs is a complex physiological process involving multiple systems,genes and signaling pathways such as Notch,ERK and Hedgehog.In this article the composition and transduction mechanisms of these crucial signaling pathways in the osteogenic differentiation of BMSCs were reviewed based on the analysis of domestic and foreign literature so as to reveal essential molecular targets and promote new ideas for gene-assisted therapeutic approaches in treating osteoporosis and bone defects.
