针刺内关穴抑制异丙肾上腺素诱导小鼠心肌肥厚的研究

摘　　要：目的:观察针刺内关穴对心肌肥厚小鼠心肌组织病理结构的影响。方法:60只雄性C57BL/6小鼠随机分为空白组、模型组、针刺组、假针刺组、药物组,每组12只。除空白组小鼠外,每只小鼠皮下注射异丙肾上腺素(ISO)25 mg·kg~(-1)·d~(-1)制备心肌肥厚模型,连续14 d。造模结束后,针刺组小鼠给予针刺双侧内关穴治疗,30 min/次;假针刺组选择小鼠尾部某一位置(不对应于任何传统穴位)进行针刺,30 min/次;药物组给予10 mg/kg普萘洛尔灌胃,1次/d,各组均治疗14 d。治疗结束后测量各组小鼠心率,超声心动图检测LVAWd、LVPWd、LVAWs、LVPWs、射血分数(EF,%);心重比(计算心脏重量/胫骨长度比值);HE染色和Masson染色观察心肌病理结构改变。结果:与空白组比较,模型组和假针刺组小鼠心率、LVAWs、LVPWs、LVPWd、LVAWd、心重比均显著升高(P<0.01,P<0.05);与模型组比较,针刺组和药物组小鼠心率、LVAWs、LVAWd、LVPWd、心重比均显著降低(P<0.01,P<0.05)。通过HE染色与Masson染色观察心肌病理形态,均可见针刺组和药物组较模型组心肌损伤程度较轻。结论:针刺内关穴能够减慢心率,抑制心肌纤维化,减轻炎症反应,改善心肌肥厚,延缓心肌损害。Objective:To observe the effect of acupuncture on the pathological structure of myocardial tissue in myocardial hypertrophic mice.Methods:Sixty male C57BL/6 mice were randomly divided into control group,model group,acupuncture group,sham group and medicine group,with 12 mice in each group.Mice were given subcutaneous injection of isoproterenol(ISO)(25 mg·kg-1·d-1)to prepare a model of myocardial hypertrophy for 14 consecutive days,except for the control group.After the modeling,the mice in the acupuncture group were treated with acupuncture bilateral Neiguan acupoint(PC 6)for 30 min/time.The sham group selected a certain position on the tail of the mouse(not corresponding to any traditional acupuncture point)for acupuncture,30 min/time.The medicine group was given 10 mg/kg propranolol by gavage once a day.All groups were treated for 14 days.After the end of treatment,the heart rate of mice in each group was measured,and LVAWd,LVPWd,LVAWs,LVPWs,and ejection fraction(EF,%)were detected by echocardiography.Calculate the heart-to-weight ratio(cardiac weight/tibial length ratio).HE staining and Masson staining were used to observe the pathological structural changes of myocardium.Results:Compared with the control group,the values of heart rate,LVAWs,LVPWs,LVPWd,LVAWd and cardiac weight ratio in the model group and the sham group were increased(P<0.01,P<0.05).Compared with the model group,the values of heart rate,LVAWs,LVPWd,LVAWd and heart-to-weight ratio were decreased in the acupuncture group and the medicine group(P<0.01,P<0.05).The pathological morphology of myocardium was observed by HE staining and Masson staining,and the degree of myocardial damage in the acupuncture group and the mdeicine group was milder than that in the model group.Conclusion:Acupuncture on Neiguan acupoint(PC 6)can slow down heart rate,inhibit myocardial fibrosis,improve cardiac hypertrophy and reduce myocardial damage.
