摘 要:目的 探讨南葶苈子水提液调控Fas/Fas配体(FasL)途径对心力衰竭大鼠心肌细胞的影响。方法 20只大鼠采用腹主动脉缩窄法建立心力衰竭模型,其中A组10只灌胃南葶苈子水提液10g·kg^(-1)·d^(-1),M组10只灌胃生理盐水5mL·kg^(-1)·d^(-1)。另将10只大鼠作为假手术(S)组,灌胃生理盐水5mL·kg^(-1)·d^(-1)。6周后,比较三组大鼠体重、心脏重量、心脏系数、心脏功能指标、血流动力学指标,HE染色观察心肌组织形态学变化,Western blot法检测Fas/FasL途径相关蛋白表达。结果 与S组比较,M组大鼠心脏重量和心脏系数升高(P<0.05),心肌细胞肥大,纤维组织排列错乱,细胞间隙增大,Fas/FasL途径相关蛋白表达升高(P<0.05)。与M组比较,A组大鼠心脏重量和心脏系数降低(P<0.05),心脏功能指标、血流动力学指标及心肌组织形态学变化均有改善(P<0.05),Fas/FasL途径相关蛋白表达降低(P<0.05)。结论 南葶苈子水提液通过调控Fas/FasL途径减轻心力衰竭大鼠心肌细胞的损伤,进而改善心力衰竭症状。Objective To investigate the effect of water extract of seeds of Descurainia sophia(L.) Webb ex Prantl through regulating Fas/Fas ligand(FasL) pathway on myocardial cells in rats with heart failure.Methods The heart failure model was established with abdominal aortic coarction method in 20 rats, which then were divided into two groups with 10 rats each.Group A was intragastrically given water extract of seeds of Descurainia sophia(L.) Webb ex Prantl 10 g·kg^(-1)·d^(-1) and group M was given normal saline 5 mL·kg^(-1)·d^(-1).Ten rats accepted sham operation(group S) were intragastrically given normal saline 5 mL·kg^(-1)·d^(-1).After 6 weeks, the body weight, heart weight, heart coefficient, cardiac function indicators and hemodynamic indicators were compared among the three groups.The myocardial histomorphology was observed by HE staining and Fas/FasL pathway-related protein expressions were detected by Western blot.Results Compared with group S,the heart weight and heart coefficient of rats were increased(P<0.05),cardiomyocytes were hypertrophy, fibrous tissues were disordered and cell space was enlarged, and Fas/FasL pathway-related protein expressions were increased in group M(P<0.05).Compared with group M,heart weight and heart coefficient of rats were decreased(P<0.05),cardiac function indicators, hemodynamic indicators and myocardial histomorphology were improved(P<0.05),and Fas/FasL pathway-related protein expressions were decreased in group A(P<0.05).Conclusion The water extract of seeds of Descurainia sophia(L.) Webb ex Prantl alleviates the damage of cardiomyocytes in rats with heart failure by regulating Fas/FasL pathway, therefore improving the symptoms of heart failure.
