摘 要:高原地区存在低氧、低压、低温、强紫外线等不良环境因素,严重影响着人们的身体健康和作业效能。低氧是高原环境主要的不良因素之一。大量研究表明,心血管系统对高原低氧非常敏感。高原低氧可扰乱心血管稳态,引起心肌细胞功能障碍甚至死亡,导致肺动脉高压、慢性高山病、高原性心脏病等高原性心血管疾病。低氧诱导因子是一类对细胞内氧浓度变化非常敏感的转录因子。高原低氧环境可激活低氧诱导因子,并通过调节一氧化氮、血管内皮生长因子、促红细胞生成素、转铁蛋白等相关因子,进一步调节心血管系统的生理病理活动,在高原性心血管疾病的发生发展以及预防和治疗中发挥重要的调节作用。本文综述了低氧诱导因子及其在高原性心血管疾病中作用,以及在抗高原性心血管疾病药物中,低氧诱导因子发挥作用的机制,探讨低氧诱导因子作为一种潜在的靶点在抗高原性心血管疾病药物研发中的应用价值。There are many hazards in high altitude areas,such as low oxygen,low pressure,low temperature and strong ultraviolet,which can impact human health and operational efficiency.Hypoxia is one of the main hazards.Numerous studies have shown that the cardiovascular system is very sensitive to high altitude hypoxia,which can disturb cardiovascular homeostasis and cause cardiomyocyte dysfunction,leading to pulmonary hypertension,chronic mountain sickness,high altitude heart disease and other high altitude cardiovascular diseases.Hypoxia inducible factors are a class of transcriptional factors that are highly sensitive to changes in intracellular oxygen concentrations.High altitude hypoxia can activate hypoxia inducible factors and regulate the pathophysiological activities of the cardiovascular system via regulation of nitric oxide,vascular endothelial growth factors,erythropoietin,transferrin and other related factors,thus playing a critical role in the development,prevention and treatment of cardiovascular diseases in high altitude areas.In this review,we summarize hypoxia inducible factors and their role in high altitude cardiovascular diseases,as well as the mechanisms of hypoxia inducible factors in high altitude cardiovascular drugs.The applicability of hypoxia inducible factors as a potential target in research and development of drugs against high altitude cardiovascular diseases is also explored.
