摘  要:【目的】明确龟纹瓢虫Propylea japonica胰岛素信号通路中4个关键基因序列结构和时空表达模式。【方法】基于龟纹瓢虫全基因组序列数据,选择E(sev)2B,TOR,PDK1和PI3K 4个基因全长进行结构分析,并通过qRT-PCR检测其在龟纹瓢虫雌虫不同发育时期(羽化后第1、3、5和7天)、不同组织(头、胸、腹和肠)中的表达模式。【结果】E(sev)2B、TOR、PDK1和PI3K基因表达水平均在成虫羽化后第7天显著上调(P<0.05),达到最高水平;E(sev)2B和PI3K基因在龟纹瓢虫羽化后第1、3和5天肠中表达水平均显著高于头、胸和腹部(P<0.05);TOR基因在羽化后第1天成虫的胸、腹、肠中表达水平均显著高于头部(P<0.05);PDK1基因在羽化第3天和第5天肠中表达水平最高,且显著高于头、胸和腹部(P<0.05)。【结论】E(sev)2B、TOR、PDK1和PI3K基因在龟纹瓢虫不同发育时期及不同组织中的表达量存在差异,为进一步研究胰岛素信号通路在龟纹瓢虫生殖发育中的作用奠定了基础。[Aim]To determine the sequence structure and spatiotemporal expression patterns of four key insulin signaling pathway genes in Propylea japonica.[Methods]The E(sev)2B,TOR,PDK1 and PI3K genes were identified in the genome of P.japonica and their coded amino sequences and functional domain locations analyzed.Expression profiles of these genes in different developmental stages(1st,3rd,5th and 7th day after eclosion)and tissues(head,thorax,abdomen and gut)were investigated using real-time quantitative PCR.[Results]Expression levels of all four genes peaked on the 7th day after adult emergence.Expression levels of the E(sev)2B and PI3K genes were significantly higher in the gut than in the head,thorax and abdomen on the 1st,3rd and 5th day after emergence(P<0.05).Expression levels of the TOR gene in the thorax,abdomen and gut were significantly higher than in the head on the 1st day after emergence(P<0.05).Expression of the PDK1 gene was highest in the gut on the 3rd and 5th day after emergence,significantly higher than in the head,thorax and abdomen(P<0.05).[Conclusion]Expression patterns of the E(sev)2B,TOR,PDK1 and PI3K genes differed in different developmental stages and tissues of P.japonica.This finding lays a foundation for further research on the role of the insulin signaling pathway in the reproductive development of this species.
