摘 要:目的探索炭疽杆菌噬菌体(Bacillus anthracis phage)的制备条件,生产效价更高的噬菌体。方法通过比较不同的宿主菌种类、培养方式、Ca^(2+)浓度、感染复数(multiplicity of infection,MOI)和培养时间对炭疽杆菌噬菌体产量的影响,选择能产生较高效价的炭疽杆菌噬菌体的制备条件。结果作为AP631株噬菌体的宿主菌,Pasteur II株比A16R株生产出的噬菌体效价高10倍;振荡培养比静止培养生产出的噬菌体效价高10倍;培养体系中的Ca^(2+)浓度为2 mmol/L时生产出的噬菌体效价最高为2.9×10^(9) PFU/mL;炭疽噬菌体菌体培养的最佳MOI是1∶30000;生产更高效价炭疽噬菌体的培养时间是8 h。结论本研究探索了炭疽噬菌体的制备过程中的宿主菌、培养方式、Ca^(2+)浓度、最佳MOI和培养时间对产出噬菌体效价的影响,为生产高效价噬菌体提供基础。Objective To obtain Bacillus anthracis phage with higher titer by exploring its preparation conditions.Methods By comparing the effects of different host strains,culture methods,Ca^(2+)concentration,MOI and culture time on the production of Bacillus anthracis phage,the best preparation conditions for obtaining higher titer Bacillus anthracis phage were determined.Results Using Pasteur II as the host strain,the titer was 10 times higher than A16R,and oscillating culture also resulted in a 10-fold increase in phage titer compared to static culture.Additionally,the highest phage titer(2.9×10^(9) PFU/mL)was achieved with the Ca^(2+)concentration of 2 mmol/L in the culture system.1∶30000 and 8 h were also considered to be the best MOI and culture time,which were more efficient and effective for the production of Bacillus anthracis phage.Conclusion This study investigated the effects of host bacteria,culture mode,Ca^(2+)concentration,optimal MOI,and culture time on the production of phage titer,these findings provide a basis for producing highly potent Bacillus anthracis phage.
