摘  要:目的 建立并评价腹泻型肠易激综合征(irritable bowel syndrome with diarrhea, IBS-D)内脏高敏感小鼠模型。方法 采用不同剂量的鼠类柠檬酸杆菌(Citrobacter rodentium,CR)灌胃联合慢性避水应激(water avoidance stress, WAS)制备IBS-D内脏高敏感小鼠模型。将60只C57BL/6J小鼠随机分为正常对照组(NC组)和模型组。模型组按不同CR剂量(0.12、0.15、0.18、0.21、0.30ml)联合WAS建立。造模后,比较小鼠粪便性状评分、粪便含水量、全肠道转运时间、结肠转运时间、腹部撤退反射(abdominal withdrawal reflex, AWR)评分,观察结肠组织病理改变。结果 与NC组比较,模型组小鼠粪便稀溏质软、粪便含水量升高、全肠道转运时间减少(P<0.05)、结肠转运时间明显减少(P<0.005)、AWR评分3分阈值注水量减少。且CR剂量0.21ml和0.30ml模型组小鼠粪便边缘模糊不清呈糊状、AWR评分3分阈值注水量明显减少(P<0.01)、结肠组织细胞排列紊乱、炎性细胞浸润、隐窝增生、皱襞变短。结论 采用CR灌胃联合WAS能够成功建立IBS-D内脏高敏感小鼠模型。结合模型各项评价结果,推荐使用0.21ml CR剂量灌胃造模。Objective To establish and evaluate a mouse model of visceral hypersensitivity in diarrhea-predominant irritable bowel syndrome(IBS-D).Methods A mouse model of IBS-D visceral hypersensitivity was established by gavage of different doses of Citrobacter rodentium(CR)combined with chronic water avoidance stress(WAS).Sixty C57BL/6J mice were randomly divided into the normal control group(NC group)and the model group.The model group was established by different doses of CR(0.12ml,0.15ml,0.18ml,0.21ml,0.30ml)combined with WAS.After modeling,the fecal trait score,fecal water content,total intestinal transit time,colonic transit time,abdominal withdrawal reflex(AWR)score,and pathological changes of colon tissue were observed.Results Compared with the NC group,the mice in the model group had soft stool,increased fecal water content,reduced total intestinal transit time(P<0.05),significantly reduced colonic transit time(P<0.005),and reduced threshold water injection of AWR score 3.In addition,in the 0.21ml and 0.30ml CR groups,the edge of feces was blurred and mush,the threshold of AWR score 3 was significantly reduced(P<0.01),the colon tissue cells were arranged disorderly,inflammatory cell infiltration,crypt hyperplasia,and the plica was shortened.Conclusion The visceral hypersensitivity mouse model of IBS-D can be successfully established by gavage of CR combined with WAS.According to the evaluation results of the model,it is recommended to use 0.21ml CR dose by gavage to establish the model.
