摘 要:目的:基于网络药理学方法结合实验验证,研究参桑茶治疗慢性疲劳综合征的作用机制。方法:利用TCMSP数据库筛选出参桑茶主要活性成分及对应靶点基因;通过DiGSeE数据库获得慢性疲劳综合征靶点基因;利用Cytoscape3.7.0软件筛选参桑茶治疗慢性疲劳综合征的关键靶点,并利用插件进行富集分析。50只雄性BALB/c小鼠随机分为5组,每组10只,分别为对照组(N)、模型组(M)、低剂量组(D1)、中剂量组(D2)、高剂量组(D3),分别进行冷水游泳、疲劳跑步、疲劳转棒和睡眠剥夺,每天1次,持续4周,第三周开始给药。取肝脏和肌肉组织,用HE染色法观察组织切片,ELISA法检测糖原、ATP、LDH、BUN含量,免疫荧光方法检测NF-κB、TGF-β蛋白表达水平。结果:富集发现核心靶点与NF-κB与TGF-β信号转导密切相关。观察HE、免疫荧光染色结果发现,用参桑茶处理后小鼠病理情况有改善,小鼠肝脏和肌肉组织中NF-κB、TGF-β表达减少。结论:参桑茶具有抗疲劳作用,分子机制可能与NF-κB、TGF-β通路有关。Objective:This study investigated the mechanism of anti-fatigue of ginseng mulberry tea based on a network pharmacology combined with experimental validation.Methods:The main active ingredients and corresponding target genes of ginseng mulberry tea were screened using the TCMSP database.The target genes of chronic fatigue syndrome were obtained by DiGSeE database.The key targets of ginseng mulberry tea for the treatment of chronic fatigue syndrome were screened using Cytoscape 3.7.0 software and enriched with plug-in analysis.Fifty male BALB/c mice were randomly divided into five groups of control(N),model(M),low-dose(D1),medium-dose(D2)and high-dose(D3)groups,10 mice each group.The mice were subjected to cold water swimming,fatigue running,fatigue bar spinning and sleep deprivation once a day for four weeks,with dosing starting at the third week.Liver and muscle tissues were collected,and tissue sections were observed by HE staining,and the contents of glycogen,ATP,LDH,and BUN were detected by ELISA,and the expression levels of NF-κB and TGF-βproteins were detected by immunofluorescence.Results:The enrichment revealed that the core targets were closely related to NF-κB and TGF-βsignal transduction.The results of HE and immunofluorescence staining showed that the pathological condition of mice treated with ginseng mulberry tea was improved,and the expressions of NF-κB and TGF-βin liver and muscle tissues of mice was reduced.Conclusion:The ginseng mulberry tea has anti-fatigue effect,and the molecular mechanism may be related to NF-κB and TGF-βpathways.
