摘 要:目的探究miR-192-5p及肿瘤相关巨噬细胞(tumor-associated macrophages,TAMs)源性外泌体对人子宫内膜癌(endometrial cancer,EC)细胞增殖及成瘤性的影响。方法采用TAMs、阴性模拟物NC转染的TAMs以及miR-192-5p转染的TAMs源性外泌体刺激子宫内膜癌细胞,并分别注入裸小鼠体内,进行肿瘤细胞移植瘤实验。然后测量各组移植瘤的体积,分别采用RT-PCR和Western blot检测肿瘤组织内miR-192-5p、相关激酶(IRAK1)和下游核因子-κB(NF-κB)表达情况,评估上调miR-192-5p的表达水平对EC细胞增殖及IRAK1和NF-κB表达水平的影响。结果TAMs源性外泌体促进肿瘤生长,miR-192-5p过表达可明显缩小肿瘤体积;miR-192-5p过表达抑制了肿瘤组织中IRAK1和NF-κB的表达。结论上调TAMs中的miR-192-5p的表达水平可能通过IRA K1和NF-κB信号通路抑制EC细胞的增殖及成瘤性。Objective The effects of miR-192-5p and tumor-associated macrophages(TAMs)on the proliferation and tumorigenesis of human endometrial cancer(EC)cells were investigated through xenograft experiments in nude mice,and the factors affecting the occurrence and development of endometrial cancer were explored.Methods Endometrial cancer cells were stimulated by TAMs,TAMs transfected with negative mimic NC and miR-192-5p transfected with TAMs-derived exosomes,which were injected into nude mice for tumor cell xenograft experiments,and then the volume of transplants in each group was measured,and miR-192-5p was detected in tumor tissues by RT-PCR and Western blot,respectively.IRAK1 and NF-kB expression,and the effect of up-regulating miR-192-5p expression levels on human endometrial cancer cell proliferation and IRAK1 and NF-kB expression levels was evaluated.Results TAMs-derived exosomes promote tumor growth,and miR-192-5p overexpression can significantly reduce tumor volume;overexpression of miR-192-5p inhibited the expression of IRAK1 and NF-kB in tumor tissues.Conclusion Up-regulation of miR-192-5p expression in tumor-associated macrophages may inhibit the proliferation and tumorigenicity of human endometrial cancer cells by inhibiting the IRAK1 and NF-kB signaling pathway.
