摘  要:目的探讨慢性砷中毒对骨性关节炎患者软骨内氧化应激水平的影响。方法本次研究共纳入42例患者,从软骨中检测出砷的样本中随机双盲抽取21例患者纳入AS-OA组,从未检测出砷的样本中随机双盲抽取21例患者纳入OA组。使用原子荧光光谱法检测样本中砷元素水平,并使用紫外-可见分光光度法检测SOD水平,硫代巴比妥酸(TBA)法检测MDA水平,分光光度法检测H_(2)O_(2)水平。比较AS-OA组和OA组的砷水平和氧化应激标志物水平,并进行相关性分析。结果AS-OA组的超氧化物歧化酶(SOD)、H_(2)O_(2)水平均高于OA组,差异具有统计学意义(P<0.05)。AS-OA组的丙二醛(MDA)水平低于OA组,但差异无统计学意义(P>0.05)。AS-OA组砷水平与SOD、MDA、H_(2)O_(2)水平之间差异无统计学意义(P>0.05)。结论慢性砷中毒可以影响软骨细胞内的氧化应激,增加了H_(2)O_(2)的表达。同时砷可能刺激SOD的表达。Objective To explore the effect of chronic arsenic poisoning on the level of oxidative stress in chondrocytes by detecting the levels of arsenic and oxidative stress markers in cartilage of osteoarthritis patients.Methods A total of 42 participants were enrolled in this study,and 21 participants who were randomly double-blind sampled from samples with detectable arsenic in cartilage were enrolled into the AS-OA group,and 21 participants who were randomly double-blind sampled from samples without detectable arsenic in cartilage were enrolled into the OA group.Atomic fluorescence spectrometry was used to detect the level of arsenic in the samples,and UV-vis spectrophotometry was used to detect the level of SOD,TBA method was used to detect the level of MDA,and spectrophotometry was used to detect the level of H_(2)O_(2).SPSS software was used for data analysis,and the levels of arsenic and oxidative stress markers SOD,MDA and H_(2)O_(2) were compared between AS-OA group and OA group,and the correlation analysis was performed.Results The SOD and H_(2)O_(2) levels in AS-OA group were higher than those in OA group,and the differences were statistically significant(P<0.05).The level of MDA in AS-OA group was lower than that in OA group,but the difference was not statistically significant(P>0.05).There was no significant correlation between arsenic level and SOD,MDA,H_(2)O_(2) levels in AS-OA group(P>0.05).Conclusion Chronic arsenic poisoning can affect the oxidative stress in chondrocytes and increase the expression of H_(2)O_(2).
