探讨MIAT通过NF-KB/IL-6/STAT3通路影响心梗后交感神经重构

摘  要:目的探讨MIAT通过NF-KB/IL-6/STAT3通路对MI后交感神经重构的影响,为临床MI后心源性猝死防治提供新的理论依据和治疗靶点。方法选取SPF级雄性Sprague-Dawley(SD)大鼠24只,根据实验需求随机分为Sham组、AMI组、AMI+shMIAT组和AMI+shControl组,每组各6只。AMI组和AMI+shMIAT组、AMI+shControl组大鼠行冠状动脉左前降支结扎术。Sham组的大鼠则接受相同的手术程序,但不结扎冠状动脉。分组干预处理后,对比各组大鼠MIAT、NGF、TNF-α、IL-1β、IL-6、NF-kB、p-P65、p-IkBa、p-JAK2、p-STAT3、JAK2和STAT3的表达水平。结果单因素方差分析结果显示,各组大鼠MIAT、NGF、TNF-α、IL-1β、IL-6、NF-kB、p-P65、p-IkBa、p-JAK2、p-STAT3、JAK2、STAT3表达水平比较差异有统计学意义(P<0.05)。两两比较结果显示,AMI组、AMI+shControl组大鼠MIAT、NGF、TNF-α、IL-1β、IL-6、NF-kB、p-P65、p-IkBa、p-JAK2、p-STAT3、JAK2、STAT3水平显著高于Sham组及MI+shMIAT组(P<0.05);AMI+shMIAT组高于Sham组,且MIAT、IL-6、STAT3差异显著(P<0.05)。AMI组与AMI+shControl组比较,差异无统计学意义(P>0.05)。结论心梗后表达的MIAT可能通过NF-KB/IL-6/STAT3通路促进心交感神经重构,进而恶性室性心律失常的发生,而降低MIAT的表达可抑制NF-KB/IL-6/STAT3通路,抑制心交感神经重构,降低心梗后恶性室性心律失常的发生,有效降低心源性猝死的发生。Objective To investigate the effect of MIAT on sympathetic remodeling after MI through NF-KB/IL-6/STAT 3 pathway,and to provide a new theoretical basis and therapeutic target for the prevention and treatment of sudden cardiac death after clinical MI.Methods 24 SPF male Sprague-Dawley(SD)rats were selected and randomly divided into Sham,AMI,AMI+shMIAT and AMI+shControl groups.Rats from the AMI and AMI+shMIAT and AMI+shControl groups were treated for left anterior descending coronary artery ligation.Rats in the Sham group underwent the same surgical procedure without ligation ating the coronary artery.After the group intervention treatment,the expression levels of MIAT,NGF,TNF-α,IL-1β,IL-6,NF-kB,p-P65,p-IkBa,p-JAK 2,p-STAT 3,JAK 2 and STAT 3 were compared.Results The results of one-way ANOVA showed that the expression levels of MIAT,NGF,TNF-α,IL-1β,IL-6,NF-kB,p-P65,p-IkBa,p-JAK 2,p-STAT 3,JAK 2 and STAT 3 were significantly(P<0.05).Pairwise comparisons showed that the AMI,MIAT,NGF,TNF-α,IL-1β,IL-6,NF-kB,p-P65,p-IkBa,p-JAK 2,p-STAT 3,JAK 2 and STAT 3 were significantly higher than Sham,and AMI+shMIAT group(P<0.05);AMI+shMIAT was higher than Sham group,and MIAT、IL-6 and STAT3 were significantly different(P<0.05).The AMI vs.AMI+shControl groups were not statistically significant(P>0.05).Conclusion The overexpressed MIAT may promote cardiac sympathetic remodeling through NF-KB/IL-6/STAT 3 pathway,and then the occurrence of malignant ventricular arrhythmia,while reducing the expression of MIAT can inhibit NF-KB/IL-6/STAT 3 pathway,inhibit cardiac sympathetic remodeling,reduce the occurrence of malignant ventricular arrhythmias after MI,and effectively reduce the occurrence of sudden cardiac death.
