摘  要:[目的]本文旨在研究层状复合氢氧化物(LDH)对dsRNA抑制稻瘟病的增强效果。[方法]比较层状复合氢氧化物(LDH)与不同比例dsRNA混合的负载效率,筛选合适的负载比。用不同浓度LDH处理稻瘟病菌孢子,考察LDH对稻瘟病菌孢子发育的影响。使用不同浓度的dsRNA-LDH处理稻瘟病菌孢子,检测dsRNA-LDH对稻瘟病菌孢子附着胞形成的抑制作用。使用稻瘟病菌侵染水稻,考察LDH增强dsRNA防治稻瘟病的效果。[结果]在100 ng·μL^(-1)浓度条件下LDH不仅不影响稻瘟病菌孢子发育,而且还可以增强dsRNA稳定性及其对稻瘟病菌孢子附着胞形成的抑制作用。与清水处理孢子的对照组相比,dsRNA-LDH处理组的水稻发病症状明显减弱。[结论]LDH可以明显增强dsRNA对稻瘟病的防治效果。dsRNA-LDH具有开发成为高效绿色环保RNAi药剂的潜力和价值。[Objectives]The study aimed to investigate the enhanced effect of layered double hydroxide(LDH)on dsRNA inhibition of rice blast disease.[Methods]The loading efficiency of LDH mixed with different ratios of dsRNA was compared,and the suitable loading ratio was screened.Different concentrations of LDH were used to treat the Magnaporthe oryzae spores to examine the impact of LDH on the development of spores.Different concentrations of dsRNA-LDH were used to treat M.oryzae spores to assess the inhibitory effect of dsRNA-LDH on the appressorium formation.Rice plants were infected with M.oryzae to examine whether LDH could enhance the efficacy of dsRNA against rice blast or not.[Results]At 100 ng·μL^(-1) concentration,LDH did not affect M.oryzae spore development,but LDH could enhance the dsRNA stability and its inhibitory effect on the formation of M.oryzae spore appressorium.Compared with the control group,the lesion area was significantly reduced on the leaf surface treated by dsRNA-LDH.[Conclusions]LDH can substantially enhance the inhibitory effect of dsRNA.dsRNA-LDH possesses the potential and value to be developed into a highly efficient and green RNAi fungicide.
