摘 要:目的:优化酒蒸王不留行炮制工艺,初步研究酒蒸王不留行治疗脂溢性皮炎的作用机制。方法:建立王不留行黄酮苷和刺桐碱的高效液相色谱(HPLC)检测方法,使用响应曲面法考察料液比、闷润时间、蒸制时间对酒蒸王不留行的影响,优化炮制工艺。建立脂溢性皮炎大鼠模型,通过观察苏木精-伊红(HE)染色病理切片和检测大鼠血清中γ干扰素(IFN-γ)、白细胞介素-2(IL-2)、白细胞介素-4(IL-4)、白细胞介素-10(IL-10)含量,探讨酒蒸王不留行治疗脂溢性皮炎的可能机制。结果:经响应曲面试验,最优酒蒸王不留行炮制工艺为:料液比25 kg/100 kg(辅料/药材),闷润时间2.4 h,蒸制时间2.4 h。药效方面:同模型组大鼠比较,阳性药物组和酒蒸组大鼠皮损评分差异有统计学意义(P<0.01)。HE染色病理切片发现,酒蒸王不留行可以修复并改善脂溢性皮炎模型表皮层和真皮层的细胞坏死,亦可以减缓由脂溢性皮炎引起的淋巴细胞浸润以及结缔组织增生的症状,阳性药物组在治疗时会引起表皮层增厚。此外,阳性药物组和酒蒸组均能提高模型大鼠血清中IL-2、IFN-γ水平的同时,降低血清中IL-4、IL-10的水平。结论:确定了酒蒸王不留行的最佳炮制工艺,且酒蒸王不留行能在一定程度上缓解脂溢性皮炎,其机制可能是通过影响辅助性T细胞1/辅助性T细胞2(Th1/Th2)平衡实现的。Objective:To optimize the processing method of wine-steamed Vaccariae Semen and preliminarily investigate its mechanism of action in treating seborrheic dermatitis.Methods:A high-performance liquid chromatography(HPLC)method was established to detect flavonoid glycosides and erythraline in Vaccariae Semen.The response surface methodology was used to examine the effects of material-to-liquid ratio,soaking time,and steaming time to optimize the processing method.A rat model of seborrheic dermatitis was established,and the potential mechanism of wine-steamed Vaccariae Semen in the treatment of seborrheic dermatitis was explored by observing hematoxylin-eosin(HE)stained pathological sections and measuring serum levels of interferon-γ(IFN-γ),interleukin-2(IL-2),interleukin-4(IL-4),and interleukin-10(IL-10).Results:Through response surface experiments,the optimal processing conditions for wine-steamed Vaccariae Semen were determined to be a material-to-liquid ratio of 25 kg/100 kg(auxiliary material/herb),a soaking time of 2.4 h,and a steaming time of 2.4 h.In terms of efficacy,compared to the model group,significant differences in skin lesion scores were observed in both the positive control group and the wine-steamed group(P<0.01).HE staining revealed that wine-steamed Vaccariae Semen could repair and improve necrosis in the epidermal and dermal layers of the seborrheic dermatitis model,as well as reduce lymphocyte infiltration and connective tissue hyperplasia caused by seborrheic dermatitis.However,the positive control group caused epidermal thickening during treatment.Additionally,both the positive control group and the wine-steamed group increased serum levels of IL-2 and IFN-γ,while reducing serum levels of IL-4 and IL-10 in the model rats.Conclusion:The optimal processing method for wine-steamed Vaccariae Semen was determined,and wine-steamed Vaccariae Semen can alleviate seborrheic dermatitis to a certain extent.Its mechanism may be related to regulating the balance between T-helper cell 1(Th1)and T-helper
