摘  要:为了研究梅花鹿(Cervus nippon)PIM1基因的结构和功能,利用RT-PCR技术扩增梅花鹿PIM1基因,获得该基因的cDNA序列,对PIM1基因及其蛋白的基本性质进行生物信息学分析,并根据PIM1蛋白的同源序列对比结果构建系统进化树,同时对PIM1基因在鹿茸生长前、中、后期顶端茸皮组织中的相对表达量进行检测。结果表明,梅花鹿PIM1基因的CDS序列全长942 bp,共编码313个氨基酸;PIM1蛋白是一种亲水性较强且无信号肽的不稳定蛋白,主要定位在细胞质和细胞核;同源性分析可知,梅花鹿PIM1蛋白与白尾鹿、黇鹿、马鹿的PIM1蛋白氨基酸序列相似度高达98.50%以上;PIM1蛋白的二级结构由43.45%的无规则卷曲、36.10%的α-螺旋、12.78%的延伸链以及7.67%的β-转角组成;实时荧光定量RT-PCR结果表明,PIM1基因在鹿茸生长后期的表达量极显著高于生长前期(P<0.01)和生长中期(P<0.01)。说明PIM1基因可能在梅花鹿鹿茸生长后期起着促进骨化的作用。In order to study the PIM1 gene structure and function,the PIM1 gene of Cervus nippon was cloned by RT-PCR,and the cDNA sequence of the gene was obtained,the basic properties of the PIM1 gene and its protein were analyzed with bioinformatics methods,and a phylogenetic tree was constructed according to the results of the comparison from the PIM1 protein homologous sequences.The relative expression of the PIM1 gene was detected at the pre-growth,mid-growth,and late-growth stages of the apical antler skin tissues.The results showed that the PIM1 gene of Sika deer was successfully amplified by RT-PCR,which was 942 bp in length and encoded 313 amino acids.The PIM1 protein was unstable and had high hydrophilicity and no signal peptide,which was mainly localized in the cytoplasm and nucleus.Homology analysis showed that the amino acid sequence similarity between the PIM1 protein of Cervus nippon and the PIM1 proteins of Odocoileus virginianus,Dama dama,Cervus canadensis were more than 98.50%.The secondary structure of the PIM1 protein consisted of 43.45%irregular convolutions,36.10%α-helix,12.78%elongation and 7.67%β-turn.According to the results of real-time RT-PCR,the expression of PIM1 gene in late-growth was significantly higher than that in pre-growth and mid-growth.It suggests that the PIM1 gene may play a role in promo-ting ossification during the later stages of antler growth in Cervus nippon.
