摘 要:本研究旨在探究KCNK3基因对不同年龄段藏绵羊睾丸可能的生物学作用。运用荧光定量PCR技术和免疫组织荧光分别检测KCNK3基因在不同发育阶段(3月龄、1岁和3岁)藏绵羊睾丸组织的表达与定位,并运用MiRWalk和TargetScan两个数据库预测与KCNK3基因具有潜在靶向调控关系的miRNAs。结果显示,KCNK3基因的表达随着藏绵羊年龄的增长呈下降趋势;免疫荧光检测结果表明,KCNK3阳性信号主要分布在各年龄段藏绵羊睾丸的精原细胞以及3月龄的生殖母细胞中;生物信息学分析结果显示,miR-296-3p和miR-331-3p与KCNK3基因的表达均呈相反趋势。综上,KCNK3基因可能通过参与精原细胞的生理活动,进而影响藏绵羊的睾丸发育和精子发生,并且KCNK3基因对藏绵羊睾丸的作用可能受miR-296-3p和miR-331-3p的调控。The aim of this study is to investigate the potential biological roles of the KCNK3 gene in the testes of Tibetan sheep of different age groups.Fluorescence quantitative PCR technology and immunofluorescence were employed to asses the expression and localization of the KCNK3 gene in the testicular tissues of Tibetan sheep at different developmental stages(age of 3 months,1 year,3 years).The MiRWalk and TargetScan databases were utilized to predict miRNAs that may have potential regulatory relationships with the KCNK3 gene.The results showed that the expression of the KCNK3 gene decreased with the age of Tibetan sheep.Immunofluorescence analysis revealed that KCNK3 positive signals were mainly distributed in the spermatogonial cells of Tibetan sheep testes at all ages and in the gonocyte at the age of 3 months.Bioinformatics analysis indicated that miR-296-3p and miR-331-3p both exhi-bited an expression trend opposite to that of the KCNK3 gene.In summary,the KCNK3 gene may influence the development and spermatogenesis of Tibetan sheep testes by participating in the physiological activities of spermatogonial cells.Additionally,the effect of the KCNK3 gene on the testes of Tibetan sheep may be regulated by miR-296-3p and miR-331-3p.
