八宝景天及香叶天竺葵混合制剂的抗炎及促愈合效果研究

摘　　要：目的:研究传统中药八宝景天及香叶天竺葵纯化提取物的主要活性成分及其对小鼠创口的抗炎及促愈合作用。方法:将八宝景天及香叶天竺葵进行纯化,使用吸光度检测及薄层硅胶板对纯化提取物中活性物质的定性及定量进行检测。取36只雄性Balb/c小鼠,随机选取18只用于皮肤刺激实验和过敏激发实验,6只用于ELISA实验,将剩余12只随机分为对照组、生理盐水组、碘伏组及实验组,每组3只。在小鼠背部脱毛后均剪出1个大小形状相似的1~1.5 cm切口,每日予以蒸馏水、生理盐水、碘伏及八宝样液喷涂,并记录小鼠伤口愈合情况。最后,在志愿者手背喷涂所制备的混合药剂,以验证其安全性。结果:经吸光度检测及薄层硅胶板验证,八宝景天及香叶天竺葵活性成分为黄酮及类黄酮物质。刺激性及过敏激发实验中未出现异常,小鼠未出现应激反应。致敏部位皮肤苏木精-伊红染色法(HE)染色无异常。小鼠创口实验中,实验组愈合速度明显较快,差异有统计学意义(P<0.05)。志愿者喷涂后喷涂部位未出现明显异常。结论:八宝景天及香叶天竺葵纯化提取物的混合制剂具有一定的抗炎及促进伤口愈合的作用。Objective:To study the main active components of purified extracts from traditional Chinese herbs Babaojingtian and Xiangyetianzhukui and their anti-inflammatory and healing effects on wounds in mice.Methods:The Babaojingtian and Xiangyetianzhukui were purified.The active substances in the purified extracts were quantitatively and qualitatively detected by absorbance detection and thin layer silica gel plate.36 male Balb/c mice were selected.18 were randomly selected for skin irritation experiment and allergy provocation experiment,and 6 were randomly selected for ELISA test.The remaining 12 mice were randomly divided into control group,normal saline group,iodophor group and experimental group,with 3 mice in each group.After hair removal,a 1-1.5 cm incision with similar size and shape was cut out on the back of the mice,and distilled water,normal saline,iodophor,and Babao sample solution were sprayed every day.The wound healing of the mice was recorded.Finally,the prepared mixture was sprayed on the back of the volunteers′hands to verify its safety.Results:The absorbance test and thin-layer silica gel plate confirmed that the active ingredients of Babaojingtian and Xiangyetianzhukui were flavone and flavonoids.The stimulation and allergic stimulation tests showed no abnormalities,and no stress response was observed in the mice.Hematoxylin-eosin(HE)staining of the sensitized skin showed no abnormality.In the mouse wound test,the healing speed of the experimental group was significantly faster,and the difference was statistically significant(P<0.05).There was no obvious abnormality in the spraying site of volunteers after spraying.Conclu⁃sion:The mixed preparation of the purified extracts of Babaojingtian and Xiangyetianzhukui has a certain effect of antiinflammatory and promoting wound healing.
