脾虚泄泻模型大鼠肠组织结构及分泌型免疫球蛋白A含量变化研究

摘  要:为研究脾虚泄泻大鼠肠道结构和肠道分泌型免疫球蛋白A的变化,试验选取24只健康SD大鼠,雌雄各半,随机分为脾虚泄泻模型组和空白对照组,在不同阶段采用“饮食失调+苦寒泻下”的方法人工复制大鼠脾虚泄泻模型,连续14 d监测大鼠的采食量、饮水量、精神状态、粪便等临床特征变化,记录第7天和第14天时大鼠的空腹体质量,采集小肠组织进行结构观察,并测定盲肠内容物中分泌型免疫球蛋白A的含量。结果表明,与空白对照组相比,模型组大鼠在试验期间出现粪便稀溏、食欲下降、消瘦、蜷缩扎堆、精神萎靡等临床症状;与空白对照组相比,模型组大鼠的体质量显著降低(P<0.05);模型组大鼠十二指肠绒毛高度,空肠隐窝深度、绒毛高度、绒毛宽度,回肠隐窝深度和绒毛宽度均显著低于空白对照组(P<0.05);模型组大鼠十二指肠隐窝深度、绒毛宽度及回肠绒毛高度均极显著低于空白对照组(P<0.01);模型组大鼠小肠不同段的绒毛高度与隐窝深度比值均低于空白对照组,但差异不显著(P>0.05);模型组大鼠盲肠内容物中sIgA含量极显著低于空白对照组(P<0.01)。结果提示,本试验成功建立了脾虚泄泻大鼠模型,脾虚泄泻可损伤大鼠小肠黏膜结构并降低盲肠中sIgA含量。The purpose was to analyze the changes of intestinal structure and sIgA content in model rats caused by experimental spleen deficiency diarrhea syndrome.This experiment artificially replicated the spleen deficiency diarrhea model using the method of“diet imbalance+bitter cold diarrhea”.24 healthy SD rats were randomly divided into a spleen deficiency diarrhea model group and a blank control group.Modeling was conducted in stages according to different conditions for 14 days.Clinical characterization changes such as food and water intake,mental state,and feces of rats were monitored daily.Fasting weight was recorded on the 7th and 14th day,and small intestine tissue was collected for structural observation.The content of secreted immunoglobulin A in the contents of the cecum was measured.Compared with the blank group,the model group rats exhibited clinical symptoms such as loose stools,decreased food intake and emaciation,curling up and piling up,and mental fatigue during the experimental period.The body weight of the model group rats was significantly lower than that of the blank group(P<0.05).The height of duodenal villi and jejunal villi,the depth of jejunal crypt and ileal crypt,and the width of jejunal villi and ileal villi in the model group were significantly lower than those in the blank group(P<0.05).The depth of duodenal crypt,the width of duodenal villus,and the height of ileal villus in the model group were extremely significantly lower than those in the control group(P<0.01).The ratio of villus height to crypt depth in different segments of the small intestine in the model group was lower than that in the blank group,but there was no significant difference(P>0.05).The sIgA content in the cecal contents of the model group rats was extremely significantly lower than that of the blank group(P<0.01).It is concluded that rat model of spleen deficiency diarrhea is successfully established in this experiment,which can damage the structure of the small intestinal mucosa and reduce the content of sIgA in t
