摘  要:目的:观察神阙灸对慢性疲劳综合征(CFS)模型大鼠生物钟基因及血清炎症因子表达的影响,探讨其作用机制。方法:21只SPF级Wistar大鼠随机分为对照组、模型组和神阙灸组,每组7只。除对照组外,模型组和神阙灸组采用通宵光照、慢性束缚、冷水游泳的方法制备CFS大鼠模型。造模成功后,神阙灸组每日给予5 min神阙灸治疗,7 d为1个疗程,共治疗4个疗程。采用行为学方法(包括一般行为学观察、鼠尾悬挂实验、力竭游泳实验)评价各组大鼠的营养状态和运动能力;采用qRT-PCR检测各组生物钟基因Clock、Bmal1 mRNA表达水平;采用ELISA法检测各组大鼠血清中白细胞介素6(IL-6)和肿瘤坏死因子-α(TNF-α)的含量变化。结果:与对照组比较,模型组大鼠体质量明显下降、力竭游泳时间明显缩短、鼠尾悬挂时间显著增加(P<0.0001),大鼠下丘脑的Bmal1 mRNA表达水平显著降低(P<0.05);血清IL-6、TNF-α含量明显升高(P<0.0001,P<0.001)。与模型组比较,神阙灸组大鼠体质量显著增加、力竭游泳时间显著延长、鼠尾悬挂时间明显缩短(P<0.05,P<0.0001),大鼠下丘脑Bmal1 mRNA表达水平明显升高(P<0.01),血清IL-6、TNF-α含量显著降低(P<0.0001)。治疗后,各组大鼠下丘脑Clock mRNA表达水平比较,差异无统计学意义。结论:神阙灸治疗可以增加CFS大鼠的体质量,增强其活力,改善睡眠,其机制可能与通过提高Bmal1 mRNA表达水平,抑制CFS大鼠体内的炎症反应相关。Objective:To observe the effects of Shenque(CV8)moxibustion on circadian clock genes and serum inflammatory factors in a rat model of Chronic Fatigue Syndrome(CFS),and to explore its mechanism of action.Methods:Twenty-one SPF-grade Wistar rats were randomly divided into three groups:Control group,Model group,and Shenque moxibustion group,with 7 rats in each group.Except for the Control group,the Model group and Shenque moxibustion group were subjected to overnight illumination,chronic restraint,and cold water swimming to establish the CFS rat model.After successful modeling,the Shenque moxibustion group received daily Shenque moxibustion treatment for 5 minutes,with 7 days constituting one course,and a total of 4 courses of treatment.Behavioral methods(including general behavioral observation,tail suspension test,and exhaustion swimming test)were used to evaluate the nutritional status and exercise ability of the rats.qRT-PCR was used to detect the mRNA expression levels of circadian clock genes Clock and Bmal1 in the rats.ELISA was used to measure serum levels of interleukin-6(IL-6)and tumor necrosis factor-alpha(TNF-α).Results:Compared to the Control group,the Model group rats showed significant reductions in body weight,shorter exhaustion swimming times,and longer tail suspension times(P<0.0001).The Bmal1 mRNA expression level in the hypothalamus was significantly reduced(P<0.05),and serum levels of IL-6 and TNF-αwere significantly increased(P<0.0001,P<0.001).Compared to the Model group,the Shenque moxibustion group rats had significantly increased body weight,longer exhaustion swimming times,and shorter tail suspension times(P<0.05,P<0.0001).The Bmal1 mRNA expression level in the hypothalamus was significantly elevated(P<0.01),and serum levels of IL-6 and TNF-αwere significantly reduced(P<0.0001).No significant differences were found in Clock mRNA expression levels in the hypothalamus among the groups after treatment.Conclusion:Shenque moxibustion treatment can increase body weight,enhance vitality,and imp
