摘 要:以二甲基亚砜和1,2-丙二醇为溶剂制备阿苯达唑亚砜(albendazole sulfoxide,ABZSO)、伊维菌素(ivermectin,IVM)复方浇泼剂;采用中心复合设计响应面法优化浇泼剂的处方;利用Franz扩散池法考察浇泼剂体外透皮性能;采用高效液相色谱法测定2种药物的渗透量。结果显示,浇泼剂的最佳配方为IVM 0.5%、ABZSO 5%、二甲基亚砜52%、丙二醇39%,余量添加无水乙醇至100%。IVM累计渗透量最高分别可达20.78μg/cm^(2),ABZSO累计渗透量最高分别可达249.02μg/cm^(2)。2种药物的体外释放模型符合一级动力学方程。1~100 mg/L ABZSO、IVM与峰面积呈良好的线性关系;2种药物的含量测定方法的精密度、稳定性RSD<2%。结果表明,ABZSO复方浇泼剂的制作工艺简单,稳定性良好,易于浇泼;所建立的高效液相色谱法操作简便,准确性高,可用于浇泼剂中ABZSO和IVM的含量测定。Albendazole sulfoxide and ivermectin compound pouring agent were prepared with dimethyl sulfoxide and 1,2-propanediol as solvents.The central composite design response surface method was used to optimize the formula of pouring agent.Franz diffusion cell method was used to investigate the transdermal performance of pouring agent in vitro.The permeation amounts of the two drugs were determined by HPLC.The best formula of pouring agent was ivermectin 0.5%,albendazole sulfoxide 5%,dimethyl sulfoxide 52%,propylene glycol 39%,and the rest was 100%anhydrous ethanol.The cumulative permeation amounts of ivermectin and albendazole sulfoxide were up to 20.78μg/cm^(2) and 249.02μg/cm^(2),respectively.The in vitro release model of the two drugs accords with the first-order kinetic equation.There is a good linear relationship between albendazole sulfoxide and ivermectin in the range of 1-100 mg/L and the peak area.The precision and stability RSD of the two methods are less than 2%.The preparation process of albendazole sulfoxide compound pouring agent is simple,stable and easy to pour.The established HPLC method is simple and accurate,and can be used for the determination of albendazole sulfoxide and ivermectin in pouring agent.
