单环刺螠对六价铬暴露响应的转录组学分析

摘  要:六价铬[hexavalent chromium,Cr(VI)]是海洋中常见的重金属污染元素之一。为探究Cr(VI)对海洋底栖动物的毒性作用,采用高通量测序技术(RNA-seq)对Cr(VI)暴露前后单环刺螠(Urechis unicinctus)体腔细胞进行转录组测序和比较分析。结果表明:Cr(VI)暴露后,筛选到1 352个差异表达基因(DEGs),其中有729个基因表达上调,623个基因表达下调。GO功能注释显示,DEGs主要富集于催化活性、结合、转运体活性、信号转导和抗氧化活性等功能。451个DEGs被注释到172个KEGG信号通路中,主要富集于真核生物核糖体生物合成、醚脂代谢、高级糖基化终末产物-受体信号通路和谷胱甘肽代谢等通路。q PCR结果显示荧光定量结果与转录组测序结果一致,证实了转录组测序结果的可靠性。本研究推测Cr(VI)暴露可激活单环刺螠GSH代谢通路,与通路相关的GSH、GST、PRDX6、RRM1、γ-GT等基因在抵御Cr(VI)暴露过程中发挥重要作用,研究结果可为重金属对海洋底栖动物的毒性机制和水环境监测研究提供参考。Hexavalent chromium[Cr(Ⅵ)]is one of the common heavy metal pollutants in marine environments.To understand the toxic effects of Cr(Ⅵ)on marine benthic animals,this study employed transcriptome sequencing to analyze the coelomocytes of Urechis unicinctus before and after exposure to Cr(Ⅵ).The results showed that 1352 differentially expressed genes(DEGs)were screened in response to Cr(Ⅵ)exposure,including 729 upregulated and 623 downregulated genes.GO functional annotation revealed that DEGs were mainly enriched in catalytic activity,binding,transporter activity,signal transduction,and antioxidant activity.Furthermore,among these DEGs,the KEGG pathway analysis mapped 451 genes to 172 pathways,with substantial enrichment in eukaryotic ribosome biosynthesis,ether lipid metabolism,AGE-RAGE signaling pathway,and glutathione metabolism.The qPCR results demonstrated a consistent pattern with the transcriptome sequencing data,which confirmed the reliability of RNA-seq results.This study suggests that the GSH metabolic pathway is activated after exposure to Cr(Ⅵ),with GSH,GST,PRDX6,RRM1,andγ-GT playing key roles in resistance to Cr(Ⅵ).This study provides valuable in-sights into the toxicity mechanisms of heavy metal exposure in marine benthic organisms and offers a basis for wa-ter environment monitoring studies.
