摘 要:目的基于16S rDNA技术,探讨电针干预膝骨关节炎兔模型对其肠道菌群的影响。方法将32只大耳白兔随机分为空白组、模型组、氨糖组、电针组,每组8只。采用改良Hulth法复制膝骨关节炎兔模型,氨糖组按60 kg比例换算灌服20 mg/kg盐酸氨基葡萄糖,每日1次,连续4周;电针组取“血海”“阴陵泉”“犊鼻”进行干预,每周5次,每次20 min,连续4周。干预结束后,收集各组兔粪便样本,采用高通量测序16S rDNA技术对兔肠道菌群进行物种差异性分析。结果Alpha多样性分析显示,与空白组相比,模型组Faith_pd指数降低,Shannon指数、Goods_coverage指数增加(P<0.05)。与模型组相比,氨糖组、电针组Chao1指数、Shannon指数均升高(P<0.05)。Beta多样性分析中,PCoA分析结果显示各组内样本相对聚集,各组间样本距离较远,UPGMA聚类分析显示样本间的分枝长度短,样本相似。物种组成分析中,在门和属水平菌群丰富度比较中,与模型组相比,氨糖组、电针组部分有益菌丰度上升,部分条件致病菌丰度下降(P<0.05)。结论电针可以通过影响膝骨关节炎兔模型肠道菌群的结构和丰度缓解膝骨关节炎,其作用可能与“肠-关节”轴之间存在的双向影响有关,但电针通过调节肠道菌群治疗膝骨关节炎的具体机制还有待进一步研究。Objective Based on 16S rDNA technology,the effect of electroacupuncture on the intestinal flora of rabbit model of knee osteoarthritis was discussed.Methods 32 white rabbits were randomly divided into blank group,model group,amino sugar group and electroacupuncture group,with 8 rabbits in each group.The rabbit model of knee osteoarthritis was reproduced by improved Hulth method.The aminoglycoside group was given 20 mg/kg glucosamine hydrochloride according to the proportion of 60 kg once a day for 4 weeks.In the electroacupuncture group,“Xuehai(SP10)”“Yinlingquan(SP9)”and“Dubi(ST35)”were used for intervention,five times a week,each time for 20 minutes,for four weeks.After the intervention,the fecal samples of rabbits in each group were collected,and the species difference of rabbit intestinal flora was analyzed by highthroughput sequencing 16SrDNA technique.Results Alpha diversity analysis showed that compared with the blank group,the Faith_pd index of the model group decreased,while Shannon index and Goods_coverage index increased(P<0.05).Compared with the model group,Chao1 index and Shannon index in the amino sugar group and electroacupuncture group were all increased(P<0.05).In the Beta diversity analysis,the results of PCoA analysis show that the samples in each group are relatively clustered,and the samples in each group are far away.UPGMA cluster analysis shows that the branching length between samples is short and the samples are similar.In the analysis of species composition,compared with the model group,the abundance of some beneficial bacteria in the amino sugar group and electroacupuncture group increased,while the abundance of some conditional pathogens decreased(P<0.05).Conclusion Electroacupuncture can alleviate knee osteoarthritis by affecting the structure and abundance of intestinal flora in rabbit model of knee osteoarthritis,and its effect may be related to the two-way influence between“gut-joint”axis,but the specific mechanism of electroacupuncture in treating knee osteoarthrit
