基于静态顶空法、超声提取法和水蒸气蒸馏法的防瘟九味香囊挥发性成分提取效果分析

摘　　要：目的比较静态顶空法、超声提取法和水蒸气蒸馏法对防瘟九味香囊挥发性成分的提取效果。方法采用超声提取法和水蒸气蒸馏法制备供试品溶液,使用气相色谱质谱联用(GC-MS)法对3种提取物进行定性分析;采用峰面积归一化法确定各化合物的相对含量。结果3种提取物中共鉴定出挥发性成分51种,其中水蒸气蒸馏提取物20种,静态顶空进样样品28种,超声提取物26种,共有成分6种。结论3种提取物的挥发性成分均以龙脑类含量较高。超声提取法提取效率较高,静态顶空法可提取沸点相对较低的挥发性成分,水蒸气蒸馏法测高挥发性成分效果较好(含量较高),3种方法综合使用可全面分析防瘟九味香囊的挥发性成分。Objective To compare the extraction effect of volatile components in Fangwen Jiuwei Sachets by the static headspace,ultrasonic extraction and hydrodistillation methods.Methods The ultrasonic extraction and hydrodistillation methods were used to prepare the test solution,and the gas chromatography-mass spectrometry(GC-MS)method was used to qualitatively analyze the three extracts.The peak area normalization method was used to determine the relative content of each compound.Results A total of 51 volatile components were identified in the three extracts,including 20 from the extracts by hydrodistillation,28 from the samples injected by static headspace,26 from the extracts by ultrasonic extraction,and six were identified as the common components.Conclusion The volatile components with high content in the three extracts are all borneol.The ultrasonic extraction method has a high extraction efficiency,the static headspace method can extract the volatile components with relatively low boiling points,and the hydrodistillation method has a better determination on highly-volatile components(with higher content).The comprehensive use of the above methods can better analyze the volatile components of Fangwen Jiuwei Sachets.
