摘　　要：目的:探讨2型糖尿病(T2DM)患者外周血单个核细胞(PBMC)长链非编码核糖核酸(lncRNA)VIM反义RNA1(VIM-AS1)、lncRNA-p53诱导转录本(LINC-PINT)表达及其临床意义。方法:选取120例T2DM患者(T2DM组),另选取同期体检健康的80例志愿者(NC组)。检测并对比两组lncRNA VIM-AS1、lncRNA LINC-PINT表达及糖脂代谢、胰岛素抵抗指标。采用受试者工作特征(ROC)曲线分析lncRNA VIM-AS1、lncRNA LINC-PINT单独及联合预测T2DM发生的价值。结果:T2DM组lncRNA VIM-AS1、lncRNA LINC-PINT表达均低于NC组(P<0.05);T2DM组三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、空腹血糖(FPG)、餐后2 h血糖(2hPG)、糖化血红蛋白(HbAlc)、空腹胰岛素(FINS)、胰岛素抵抗指数(HOMA-IR)水平均高于NC组,高密度脂蛋白胆固醇(HDL-C)水平低于NC组(P<0.05);绘制ROC曲线发现,lncRNA VIM-AS1、lncRNA LINC-PINT单独及联合预测T2DM发生的曲线下面积(AUC)为0.824、0.746、0.900,联合预测价值高于各指标单独检测。结论:T2DM患者PBMC中lncRNA VIM-AS1、lncRNA LINC-PINT呈低表达,联合检测对T2DM发生具有较高的预测价值。Objective:To investigate the expression and clinical significance of long non-coding RNA(lncRNA)VIM antisense RNA1(VIM-AS1)and lncRNA-p53 induced transcript(LINC-PINT)in peripheral blood mononuclear cells(PBMC)of patients with type 2 diabetes(T2DM).Methods:120 patients with T2DM(T2DM group),and 80 volunteers(NC group)were selected during the same period.The expression of lncRNA VIM-AS1,lncRNA LINC-PINT,glucose and lipid metabolism and insulin resistance were detected and compared between two groups.The value of lncRNA VIM-AS1,lncRNA LINC-PINT alone and in combination in predicting the occurrence of T2DM were analyzed by receiver operating characteristic(ROC)curve.Results:The expression of lncRNA VIM-AS1 and lncRNA LINC-PINT in T2DM group was lower than that in NC group(P<0.05).The levels of triglyceride(TG),total cholesterol(TC),low density lipoprotein cholesterol(LDL-C),fasting plasma glucose(FPG),2 h postprandial blood glucose(2hPG),glycosylated hemoglobin(HbAlc),fasting insulin(FINS)and insulin resistance index(HOMA-IR)in T2DM group were higher than those in NC group,the level of high density lipoprotein cholesterol(HDL-C)was lower than that in NC group(P<0.05).ROC curve showed that,the area under the curve(AUC)of lncRNA VIM-AS1 and lncRNA LINC-PINT alone and in combination to predict the occurrence of T2DM were 0.824,0.746 and 0.900,respectively,the combined predictive value was higher than that of each index alone.Conclusion:The low expression of lncRNA VIM-AS1 and lncRNA LINC-PINT in PBMC of patients with T2DM.The combined detection has a high predictive value for the occurrence of T2DM.
