加压联合抗阻训练对T2DM大鼠骨骼肌胰岛素抵抗及质量的影响

摘  要:目的:探讨加压联合低强度抗阻训练对2型糖尿病(T2DM)大鼠糖代谢、骨骼肌胰岛素抵抗(PI3K/Akt/Glut4)通路及其下游P70S6K蛋白表达和质量的影响。方法:HFD饲料喂养8周,联合注射35mg/kg链脲佐菌素构建T2DM大鼠模型,造模成功大鼠均分为5组(n=9),2型糖尿病对照组(DR):不进行运动干预;单纯加压训练组(BRT):加压5 min休息5 min,4次循环;高强度抗阻训练组(HRT):50%~75%1RM;低强度抗阻训练组(LRT):35%~55%1RM;加压联合低强度抗阻训练组(BLRT):加压联合35%~55%1RM。运动后测糖代谢指标,Western blot法测PI3K/Akt/Glut4通路及P70S6K相关蛋白表达。结果:①8周后,BRT组、LRT组、BLRT组空腹血糖显著下降(P<0.05)。②与DR组比较,HRT组p-PI3K、p-Akt和Glut4显著增加(P<0.01);与BRT组比较,HRT组p-PI3K、Glut4显著增加(P<0.05)。③与DR组比较,BLRT组p-Akt、Akt/p-Akt和Glut4显著增加(P<0.01);与BRT组比较,BLRT组Glut4表达显著增加(P<0.01);与HRT组比较,BLRT组Akt/p-Akt比值显著增加(P<0.05)。④BLRT组和HRT组p-Akt、Glut4、P70S6K、p-P70S6K蛋白表达和p-P70S6K/P70S6K比值相近。结论:加压联合低强度抗阻训练能够增加p-Akt/Akt的比值,上调Glut4表达,改善T2DM大鼠骨骼肌胰岛素抵抗,降低血糖。加压联合抗阻训练对骨骼肌质量无显著影响。Objective:To investigate the effects of blood flow restriction combined with low-intensity resistance training on glucose metabolism,skeletal muscle insulin resistance(PI3K/Akt/Glut4)pathway and its downstream P70S6K protein expression and mass in T2DM rats.Methods:The typeⅡdiabetes mellitus rat model was constructed by feeding high-sugar and high-fat diet for 8 weeks and injecting 35 mg/kg streptozotocin.The successfully modeled typeⅡdiabetes mellitus rats were divided into 5 groups(n=9).TypeⅡdiabetes control group(DR):no exercise intervention;pure blood flow restriction training group(BRT):blood flow restriction for 5 minutes,rest for 5 minutes,4 cycles;high-intensity resistance training group(HRT):50%~75%1RM;low-intensity resistance training group(LRT):35%~55%1RM;blood flow restriction combined with low-intensity resistance training group(BLRT):blood flow restriction combined with 35%~55%1RM.After exercise,glucose metabolism indexes were measured,and the expression of PI3K/Akt/Glut4 pathway and P70S6K-related proteins were measured by Western blot.Results:①After 8 weeks,fasting blood glucose in BRT,LRT and BLRT groups decreased significantly(P<0.05).②Compared with DR group,p-PI3K,p-Akt and Glut4 were significantly increased in HRT group(P<0.01).Compared with BRT,p-PI3K and Glut4 in HRT group were significantly increased(P<0.05).③Compared with DR group,p-Akt,Akt/p-Akt and Glut4 were significantly increased in BLRT group(P<0.01).Compared with BRT,Glut4 expression in BLRT group was significantly increased(P<0.01).Compared with HRT,the Akt/p-Akt ratio in BLRT group was significantly increased(P<0.05).④The expression of p-Akt,Glut4,P70S6K,p-P70S6K protein and the ratio of p-P70S6K/P70S6K were similar in BLRT and HRT groups.Conclusion:Blood flow restriction combined with low-intensity resistance training increases the p-Akt/Akt ratio,upregulates Glut4 expression,and attenuates skeletal muscle insulin resistance in T2DM rats,thereby reducing blood glucose.Blood flow restriction combined with resistance
