茭白WRKY基因家族鉴定及菰黑粉菌侵染下的表达分析

摘  要:WRKY基因家族是植物最大的转录因子家族之一,在植物生长发育及逆境胁迫响应过程中起重要调控作用。基于茭白基因组数据库,鉴定WRKY基因家族并对其进行生物信息学分析,同时研究菰黑粉菌侵染下ZlWRKYs的表达模式。结果表明,茭白基因组中共鉴定出120个ZlWRKY家族基因,命名为ZlWRKY1~ZlWRKY120,分布在17条染色体上。ZlWRKYs可分为Ⅰ、Ⅱ和Ⅲ类,其中Ⅰ类有17个ZlWRKYs,Ⅱ类有72个ZlWRKYs,Ⅲ类有31个ZlWRKYs。茭白ZlWRKYs成员在进化上存在基因片段复制;启动子顺式作用元件分析表明,ZlWRKYs包含多个与激素、逆境胁迫响应和生长发育相关的元件。菰黑粉菌侵染3 h,大部分ZlWRKYs基因呈现上调表达,其中ZlWRKY33相对表达量最高。本研究为深入探讨茭白ZlWRKYs基因功能以及解析雄茭响应菰黑粉菌侵染的分子机制提供理论基础。WRKY gene family is one of the largest transcription factor families in plants and plays an important role in the regulation of plant growth and development and response to stress.In this study,WRKY gene family was identified,and bioinformatic analysis was conducted based on the genome database of Zizania latifolia.Meanwhile,the expression pattern of ZlWRKYs gene after inoculation with Ustilago esculenta was also studied.The results showed that a total of 120 ZlWRKY genes,named ZlWRKY1-ZlWRKY120,were identified in the genome of Z.latifolia,and they were distributed on chromosome 17.ZlWRKYs could be divided into Groups Ⅰ,Ⅱ and Ⅲ,of which there were 17 ZlWRKYs in Group Ⅰ,72 ZlWRKYs in Group Ⅱ and 31 ZlWRKYs in Group Ⅲ.Gene fragment duplication existed in ZlWRKYs members.The analysis of promoter cis-acting elements showed that ZlWRKYs contained several elements related to hormones,growth and development and stress response.At 3 h after inoculation with U.esculenta,most ZlWRKYs genes showed up-regulated expression,and the relative expression of ZlWRKY33 was the highest.This results provide a theoretical basis for further studying on the function of ZlWRKYs gene in Z.latifolia and the molecular mechanism of male Z.latifolia response to U.esculenta infection.
