摘 要:为了解瓦氏黄颡鱼的种质遗传状况,进而为其良种选育提供科学依据,本研究采集了我国瓦氏黄颡鱼主要养殖区(广东、四川、浙江和安徽)6个养殖群体共168尾个体,利用微卫星标记分析了其遗传多样性与遗传结构。结果显示:所有群体均有较高的遗传多样性水平(Na=6.500~8.250,Ho=0.758~0.869,He=0.729~0.806,PIC=0.672~0.762)且各群体间遗传多样性水平相近。群体间遗传距离(0.0575~0.6505)、遗传分化指数(0.0119~0.1522)及AMOVA分析(群体间遗传变异比9.93%)结果共同表明瓦氏黄颡鱼群体间存在显著遗传分化;群体间系统进化树显示所有群体具有共同的祖先型,聚为互为姊妹群的两大支,其中安徽的3个群体聚为一支,其余群体聚为另一支;利用Structure 2.3.4软件的遗传聚类分析显示最佳遗传分组数为3,其中广东、四川和浙江3个群体遗传组成相似,聚为一个遗传组,应为长江种质来源;淮南和六安两群体间遗传组成相似,聚为一个遗传组,应为淮河干流种质来源;凤台群体单独为一个遗传组,应为淮河附属阻隔湖泊-城北湖种质来源,故各遗传组间遗传组成差异明显。研究表明,6个瓦氏黄颡鱼养殖群体的遗传多样性高、遗传分化显著,具有很大选育潜力,可为瓦氏黄颡鱼的良种选育提供优良种质资源库。In order to understand the genetic status of Pelteobagrus vachelli germplasm,which provides a forceful scientific foundation for improved variety breeding of P.vachelli,a total of 168 individuals of P.vachelli were collected from major aquaculture areas(Guangdong,Sichuan,Zhejiang and Anhui Province)in China.The genetic diversity and genetic structure of 6 populations were analyzed using microsatellite markers.The results showed that all populations exhibited high levels of genetic diversity(Na=6.500−8.250,Ho=0.758−0.869,He=0.729−0.806,PIC=0.672−0.762),with similar levels of genetic diversity among them.The genetic distance between populations ranged from 0.0575 to 0.6505,the genetic differentiation index from 0.0119 to 0.1522,and the proportion of inter-population genetic variation in AMOVA was 9.93%(>0.05),which indicated that there was significant genetic differentiation among these populations.The phylogenetic tree showed that the 6 populations had a common ancestor type and were clustered into two branches of each other's sister groups,among which 3 populations from Anhui were clustered into one branch and the rest were clustered into another.The results of genetic cluster analysis using the software Structure 2.3.4 showed that the optimal number of genetic grouping was 3.The genetic composition of Guangdong,Sichuan and Zhejiang populations was similar,clustering them into one genetic group,which should be the germplasm source of Yangtze River.The genetic composition of Huainan and Lu'an populations was also similar,clustering them into another genetic group,which should be the germplasm source of the main stream of the Huai River.The Fengtai population was a single genetic group,likely representing the germplasm source of the isolated lake,Chengbei Lake,a tributary of the Huai River.There were significant differences in genetic composition among all the genetic groups.The results demonstrated that the six populations of P.vachelli had high genetic diversity and significant genetic differentiation,ind
