摘 要:为探究壳寡糖水溶液(商品名优力克)体外抗菌活性及其影响因素,本研究采用标准微量肉汤稀释联合平板计数法(简称标测法)测定了优力克对4株渔源多重耐药菌的最小抑菌浓度(minimum inhibitory concentration,MIC)和最小杀菌浓度(minimum bactericidal concentration,MBC)、采用微孔板模拟全池泼洒(简称模测法)测定了优力克对MHA可培养水生细菌的MIC和MBC、并评价了标测法的孵育温度、起始细菌浓度、MH浓度、pH和货架期对优力克MIC和MBC的影响。结果显示,标测法下,优力克对大肠杆菌E105、维氏气单胞菌AV040、副炭疽芽孢杆菌BC006和贝莱斯芽孢杆菌B14的MIC和MBC(32~64 g·m^(-3))是其临床推荐使用浓度(0.3~0.5 g·m^(-3))的数百倍;而模测法下,优力克对MHA可培养水生菌的MIC和MBC(4~8 g·m^(-3))是其临床推荐使用浓度的数十倍。标测法体系的温度(5~35℃)越高、细菌浓度(103~108 cfu·mL^(–1))越高、MH浓度(1/256~1×MH)越高、pH(5.5~9.5)越高,优力克对细菌的MIC和MBC越大(最大值是最小值的2~8倍);上述参数范围内优力克对4种测试菌的MIC在16~128 g·m^(-3)之间。优力克货架保质期至少为24个月。本研究结果提示,壳寡糖水溶液(优力克)是非常有前景的渔业水环境细菌载量的调节剂。To investigate the antibacterial activity in vitro and impact factors of chitosan oligosaccharides aqueous solution(trade name:Yulike),the standard micro broth dilution combined with plate counting method(referred to as the standard measurement method)was used to determine the minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of Yulike,micro plate simulation whole pool sprinkling(simulated measurement method)was used to determine the MIC and MBC of Yulike against MHA cultivable aquatic bacteria,and the effects of incubation temperature,initial bacterial concentration,MH concentration,and pH of the measurement system,as well as shelf life on Yulike MIC and MBC were evaluated.The results showed that,the MIC and MBC(32-64 g·m^(-3))of Yulike against Escherichia coli E105,Aeromonas veronii AV040,Bacillus paranthracis BC006,and Bacillus velezensis B14 were hundreds of times higher than the clinical recommended concentrations(0.3-0.5 g·m^(-3));the MIC and MBC values(4-8 g·m^(-3))of MHA culturable aquatic bacteria by Yulike were dozens of times higher than the clinical recommended concentrations.The higher temperature(5-35℃),bacterial concentration(103-108 cfu·mL^(-1)),MH concentration(1/256-1×MH),and pH(5.5-9.5)of the measurement system,the greater the MIC of Yulike against bacteria(the maximum value is 2-8 times the minimum value),and the MIC of Yulike against the four tested bacteria ranged from 16 g·m^(-3)to 128 g·m^(-3).The shelf life of Yulike is at least 24 months.These results suggested that Yulike is a very promising regulator of bacterial load in the fishing water environment.
