摘 要:目的探讨姜黄素对2型糖尿病大鼠心肌损伤的保护作用及机制。方法雄性SD大鼠高脂高糖饲料喂养4周后腹腔注射STZ建立2型糖尿病模型,姜黄素保护组采用姜黄素灌胃治疗12周。检测大鼠空腹血糖、心脏指数,HE染色观察心肌组织形态学改变,试剂盒测定各组大鼠心肌组织超氧化物歧化酶活性以及脂质氧化水平,Western Blot检测各组大鼠心肌组织中和mTOR信号通路相关蛋白的表达水平。结果与对照组相比,模型组大鼠心脏指数明显增加(P<0.05);心肌组织形态紊乱;超氧化物歧化酶(superoxide dismutase,SOD)活性明显下降(P<0.05);丙二醛(malondialdehyde,MDA)含量明显升高(P<0.05);PI3k/β-actin、p-AKT/AKT、mTOR/β-actin水平明显降低(P<0.05)。与模型组相比,姜黄素保护组大鼠心脏指数明显降低(P<0.05);心肌组织形态改善;SOD活性明显升高(P<0.05);MDA含量明显下降(P<0.05);PI3k/β-actin、p-AKT/AKT、mTOR/β-actin水平明显升高(P<0.05)。结论姜黄素可以通过自身的抗氧化性改善糖尿病大鼠心肌组织的PI3K/AKT/mTOR信号通路活性,发挥心肌保护作用。Objective To investigate the protective effect and mechanism of curcumin on myocardial injury in type 2 diabetes rats.Methods Male SD rats were fed with high fat and high sugar diet for 4 weeks,and then intraperitoneal injection of STZ was used to establish the model of type 2 diabetes.Curcumin protection group was treated by intragastric administration of curcumin for 12 weeks.Fasting blood glucose and cardiac index of rats were detected;HE staining was used to observe the morphological changes of myocardial tissue;the SOD activity and MDA content in the myocardial tissue of rats in each group were measured using a reagent kit;Western Blot was used to detect the expression level of myocardial tissue related proteins in each group of rats.Results Compared with the control group,the heart index of the model group rats was significantly increased(P<0.05);myocardial tissue morphology was disturbed;SOD activity was significantly decreased(P<0.05);the MDA content was significantly increased(P<0.05);the levels of PI3k/β-actin,P-Akt/AKT and mTOR/β-actin were significantly decreased(P<0.05).Compared with the model group,the cardiac index of rats in the curcumin protected group was significantly reduced(P<0.05);myocardial tissue morphology was improved;SOD activity was significantly increased(P<0.05);the MDA content was significantly decreased(P<0.05);the levels of PI3k/β-actin,P-Akt/AKT,mTOR/β-actin were significantly increased(P<0.05).Conclusion Curcumin can improve the activity of PI3K/AKT/mTOR signaling pathway in myocardial tissue of diabetes rats through its own antioxidant activity,and can play a role in myocardial protection.
