摘 要:为探究阿克苏地区硬蜱的种类及遗传进化特征,从阿克苏地区4个县(市)的羊体表共采集到1016只硬蜱。用显微镜观察硬蜱的形态结构,用聚合酶链反应(PCR)方法鉴定蜱种,选择阳性样本进行测序,同源性比对并进行系统发育分析。结果表明,共3属5种蜱,分别为亚洲璃眼蜱(Hyalomma asiaticum)509只,小亚璃眼蜱(Hyalomma anatolicum)133只,图兰扇头蜱(Rhipicephalus turanicus)246只,血红扇头蜱(Rhipicephalus sanguineus)117只及边缘革蜱(Dermacentor marginatus)11只。亚洲璃眼蜱为优势蜱种(Dr=50.10%,509/1016)。基于16S rDNA基因测序比对发现,采集的5种蜱同种间的同源性均达100%。ITS-2测序的亚洲璃眼蜱、图兰扇头蜱、血红扇头蜱的同源性达99%以上。说明阿克苏地区是蜱虫流行区,建议加强蜱的防控。In order to investigate the species and genetic evolutionary characteristics of Ixodidae in the Aksu region,a total of 1016 ticks were collected from the body surface of sheep in four counties/cities in the Aksu region.Using a microscope to observe the morphological structure of ticks,employing the polymerase chain reaction(PCR)method to identify tick species,selecting positive samples for sequencing,conducting homology alignment,and performing phylogenetic analysis.The results showed that there were a total of 3 genera and 5 species of ticks,including 509 individuals of H.asiaticum,133 individuals of H.anatolicum,246 individuals of R.turanicus,117 individuals of R.sanguineus,and 11 individuals of D.marginatus.H.asiaticum was the dominant tick species(Dr=50.10%,509/1016).Based on the alignment of 16S rDNA gene sequencing,it was found that the homology among the five collected tick species reached 100%within the same species.The homology of ITS-2 sequencing among H.asiaticum,R.turanicus,and R.sanguineus exceeds 99%.This experiment indicates that the Aksu region is an endemic area for ticks,suggesting the need to strengthen tick control measures.
