摘 要:波棱瓜子是一种传统藏药,主要用于治疗肝胆疾病,具有保肝利胆的作用。首先提取波棱瓜子总木脂素,并探究波棱瓜子总木脂素低、中、高剂量组(375、750、1500 mg/kg)对由对乙酰氨基酚(APAP)诱导的小鼠急性肝损伤的保护作用。末次给药后1 h,除空白对照组外,其余各组小鼠以350 mg/kg剂量腹腔注射APAP溶液。试验组小鼠禁食不禁水16 h后对小鼠称重后取材,观察各组肝脏大体情况和肝组织的组织病理学变化。采血并测定血清中AST、ALP和ALT的含量。结果表明,与空白对照组相比,APAP模型组肝脏有明显水肿,肝脏系数显著增大,血清AST、ALP和ALT含量均明显增高(P<0.05)。组织病理学结果显示,正常组小鼠的肝细胞形态正常。模型组小鼠的肝索结构紊乱,有大量炎症细胞,肝细胞核固缩、空泡。与APAP模型组比较,波棱瓜子总木脂素各剂量组肝脏水肿减轻,肝脏系数、AST、ALP、ALT含量显著低于模型组(P<0.05),小鼠肝脏组织的异常病理变化得到改善。说明波棱瓜子总木脂素对APAP诱导的小鼠急性肝损伤具有保护作用。The Herpetospermun caudigerm is a traditional Tibetan medicine,which is mainly used to treat liver and gallbladder diseases,and it has the function of protecting liver and gallbladder.In this study,we extracted the total lignans from Herpetospermun caudigerm,and further studied the protective effects of different doses of total lignans(375 mg/kg,750 mg/kg,1500 mg/kg)on acetaminophen(APAP)-induced acute liver injury in mice.One hour after the last administration,all mice except control group in the other groups were administered with acetaminophen(350 mg/kg)for one time by intraperitoneal injection.After the mice in the experimental group were deprived of water for 16 h,the bodies of mice were weighed and the samples were harvested,the appearance and histopathology of liver were observed.The levels of aspartate aminotransferase(AST),alkaline phosphatase(ALP)and alanine aminotransferase(ALT)in serum were measured.Compared with control group,the appearance and histopathology of liver were bad in the model groups,the liver is hepatomegaly,the liver coefficient,and the levels of AST,ALP and ALT contents in serum were also significantly increased(P<0.05).The histopathological results indicated that the morphology of liver cells in the normal group was normal,while the liver cord structure was disordered in the APAP model group,with a large number of inflammatory cells,nuclear shrinkage and vacuole.Compared with APAP model group,liver edema was alleviated and liver coefficient,AST,ALP and ALT contents were significantly decreased in the total lignans treated groups(P<0.05).Abnormal histopathological changes of liver were improved.These results indicated that the total lignans of the Herpetospermun caudigerm.play a protective role in APAP-induced acute liver injury in mice.
