摘 要:为探究乌龙茶提取物(OTE)取代醋酸双氧铀(UA)在畜牧生物样本透射电镜制样中的染色效果,选取畜牧业中3种常见的生物样本即山羊坐骨神经、酵母菌及苜蓿叶片组织为测试对象,经多聚甲醛-戊二醛PBS混合溶液预固定后制备超薄切片,采用OTE染色,再用透射电镜观察样品反差情况,并与传统UA染色法进行比较。结果显示3种生物样品的OTE单染组和双染组(OTE-Pb双染)在反差对比度、图像清晰度、结构辨识度等方面均与对应的UA单染组和双染组(UA-Pb双染)相当,甚至更佳。结果说明OTE染色在畜牧生物类样品透射电镜制样中的染色效果稳定,反差良好,而且比常规使用的UA安全性高,具有更广阔的应用前景。In order to investigate the staining effects of oolong tea extract(OTE)as a substitute for uranyl acetate(UA)in transmission electron microscopy(TEM)sample preparation,this study selected three common biological samples:goat sciatic nerve,yeast cells,and clover leaf tissue.After pre-fixation with a mixture of paraformaldehyde and glutaraldehyde in PBS,ultra-thin sections were prepared.The sections were stained with OTE,and the resulting contrast was observed using TEM,comparing them with the traditional UA staining method.The results showed that both the OTE single staining and double staining with lead(OTE-Pb double staining)groups exhibited comparable or even superior contrast,image clarity,and structural identification as compared to the corresponding UA single staining and double staining with lead(UA-Pb double staining)groups for the three biological samples.The research findings indicate that OTE staining method provides stable staining effects,good contrast,and higher safety compared to the conventional use of UA in TEM sample preparation for biological samples.Furthermore,OTE staining solution is safer than the conventional use of UA,demonstrating a broader range of potential applications.
