摘 要:目的建立IL-1β诱导的小鼠慢性阻塞性肺疾病气道重塑模型,并进行系统性评价。方法将15只C57BL/6J小鼠分为对照组、7 d组和14 d组。7、14 d组气管内注射Ad-IL-1β,对照组气管内注射Ad-LacZ。对照组14 d后取材,实验组在7、14 d分别取材,通过观察小鼠一般情况、肺功能检测、肺组织Masson染色、肺组织HE染色、外周血ELISA检测炎症因子并采用单因素方差分析进行数据比较,判断模型是否成功建立及评价该模型是否符合慢性阻塞性肺疾病气道重塑的临床特点。结果与对照组相比,7、14 d组小鼠肺功能显著下降,小气道周围大量炎症细胞浸润、胶原沉积明显、外周血中炎症因子明显增加(P<0.05)。结论采用IL-1β气管内给药可成功构建符合慢性阻塞性肺疾病临床特点的气道重塑模型。Objective To establish and systematically evaluate an IL-1β-induced airway remodeling model in mice with chronic obstructive pulmonary disease(COPD).Methods Fifteen C57BL/6J mice were divided into control group,7-days and 14-day groups.Ad-IL-1βwas injected intratracheally in the 7-day and 14-day groups,and Ad-LacZ was injected intratracheally in the control group.Samples were collected from the control group after 14 days,and samples were collected from the experimental group at 7 days and 14 days,respectively,to determine whether the model was successfully established.The general condition and pulmonary function of mice was recorded,the lung tissue was observed after Masson and HE staining,and inflammatory factors in peripheral blood was detected by ELISA.Results Compared with the control group,the pulmonary function of mice in the 7-day group and 14-day group was significantly decreased,a large number of inflammatory cells infiltrated around the small airways,collagen deposition was significant,and inflammatory factors in the peripheral blood were significantly increased(P<0.05).Conclusion The airway remodeling model of chronic obstructive pulmonary disease can be successfully established by intratracheal administration of IL-1β.
