摘 要:【目的】探究急性温度和盐度胁迫对罗氏沼虾热休克蛋白基因及抗氧化酶活性的影响,为实际生产中罗氏沼虾的健康养殖提供理论依据。【方法】采用双因素试验,设不同温度(20、24、28和32℃)和盐度(0.5‰、4.0‰、8.0‰和12.0‰),共16个处理组,将罗氏沼虾从初始温度(24℃)和盐度(0.5‰)环境转移至各处理组环境中进行急性胁迫,分别在胁迫24和48 h时采集罗氏沼虾鳃组织及肝胰腺组织,测定热休克蛋白70基因(HSP70)相对表达量、超氧化物歧化酶(SOD)活性和谷胱甘肽过氧化物酶(GPX)活性。【结果】胁迫24 h时,在相同温度下,随着盐度的升高,罗氏沼虾鳃组织和肝胰腺组织HSP70基因相对表达量均整体呈先上升后下降的变化趋势;在0.5‰和4.0‰盐度下,温度为20、28和32℃时罗氏沼虾鳃组织和肝胰腺组织HSP70基因相对表达量均高于24℃,且肝胰腺组织HSP70基因相对表达量在温度为28和32℃时显著高于20℃(P<0.05,下同)。胁迫48 h后,温度为24℃时,8.0‰和12.0‰盐度下罗氏沼虾鳃组织及肝胰腺组织HSP70基因相对表达量均显著高于0.5‰与4.0‰盐度。胁迫24 h时,在相同温度下,随着盐度的升高,罗氏沼虾腮组织SOD和GPX活性均整体呈上升趋势;温度为24℃时,随着盐度升高,肝胰腺组织SOD和GPX活性均无显著变化(P>0.05),而在其他温度下出现明显变化;在相同盐度下,随着温度的升高,罗氏沼虾鳃组织SOD和肝胰腺组织GPX活性均整体呈先下降后上升的变化趋势,腮组织GPX活性和肝胰腺组织SOD活性均整体呈上升趋势。双因素方差分析结果表明,温度、盐度及其交互作用对罗氏沼虾鳃组织和肝胰腺组织HSP70基因表达与抗氧化酶(SOD和GPX)活性均有极显著影响(P<0.01)。【结论】急性温度和盐度胁迫会引起罗氏沼虾HSP70基因相对表达量升高,且温度骤升较温度骤降更能诱导HSP70基因表达。温度和盐度骤变能引起罗氏沼�【Objective】To investigate the effects of acute temperature and salinity stress on the heat shock protein gene and antioxidant enzyme activity of Macrobrachium rosenbergii,which could provide reference basis for the healthy aquaculture of M.rosenbergii in actual production.【Method】Using two-factor test with different temperatures(20,24,28 and 32℃)and salinity(0.5‰,4.0‰,8.0‰and 12.0‰),16 treatment groups in total,transferred M.rosenbergii from the initial temperature(24℃)and salinity(0.5‰)to the environment of each treatment group for acute stress.At 24 and 48 h of stress,the gill and hepatopancreas tissues of M.rosenbergii were taken for measurement of the expression level of heat shock proteins 70 gene(HSP70)and the activities of superoxide dismutase(SOD)and glutathione peroxidase(GPX).【Result】At 24 h of stress,with the increase of salinity,the relative expression of HSP70 gene in gills and hepatopancreas increased first and then decrease;at 0.5‰and 4.0‰salinity,the relative expression of HSP70 gene in gills and hepatopancreas of M.rosenbergii at 28 and 32℃was higher than 24℃,moreover,the relative expression of HSP70 gene in hepatopancreas at 28 and 32℃was significantly higher than 20℃(P<0.05,the same below).After 48 h of stress,the relative expression of HSP70 gene at 8.0‰and 4.0‰salinity at 24℃of gill and hepatopancreas tissues in M.rosenbergii was higher than 0.5‰and 4.0‰salinity.Under the same temperature at 24 h of stress,the activities of SOD and GPX in gill tissue of M.rosenbergii increased with the increase of salinity.When temperature was 24℃,SOD and GPX activities in hepatopancreas tissue had no significant changes with the increase of salinity(P>0.05),but had ob-vious changes at other temperatures.Under the same salinity,SOD activity in gill tissue and GPX activity in hepatopancreas tissue decreased first and then increased with the increase of temperature,GPX activity in gill tissue and SOD activity in hepatopancreas tissue increased as a whole.The res
