消风宣窍汤对变应性鼻炎的治疗作用及代谢组学研究

摘　　要：目的分析变应性鼻炎小鼠血清代谢物变化,观察消风宣窍汤干预后差异代谢物变化,基于代谢组学分析消风宣窍汤的干预效果。方法选取40只小鼠,10只作为空白组,其余30只建立变应性鼻炎小鼠模型,共建模成功27只,随机分为模型组、西药组、消风宣窍汤组各9只。消风宣窍汤组给予消风宣窍汤,西药组给予孟鲁司特钠,空白组、模型组给予80mg/kg生理盐水灌胃。评估各组小鼠建模成功当天、干预后第1日、第3日、第7日行为学评分,分析消风宣窍汤对血氧样本代谢谱的影响。结果与空白组相比,模型组、西药组、消风宣窍汤组在建模成功当天、干预后第1日、第3日、第7日行为学评分较高(P<0.05),模型组、西药组、消风宣汤组建模成功当天行为学评分对比差异无统计学意义(P>0.05);与模型组相比,西药组、消风宣窍汤组干预后第1日、第3日、第7日行为学评分较低(P<0.05),西药组、消风宣窍汤组千预后第1日、第3日、第7日行为学评分对比差异不明显(P>0.05)。PCA分析结果显示,各组代谢物存在一定差异,表明变应性鼻炎改变了小鼠血清中的某些物质代谢,经消风宣窍汤、孟鲁司特钠干预后,变化代谢物有回归趋势。OPLS-DA分析结果显示,各组间分离趋势较为明显,与空白组相比,模型组血清中有46种代谢物发生显著变化,而经消风宣窍汤干预后14种差异性代谢物发生逆转。通路影响值分析结果显示,共得到9条与变应性鼻炎相关的代谢通路,其中4条与消风宣窍汤治疗变应性鼻炎相关。结论变应性鼻炎小鼠血清中多种代谢物处于异常状态,消风宣窍汤可调节差异代谢物,使其回归正常状态,最终改善小鼠行为学状态。Objective:To analyze the changes of serum metabolites in rats with allergic rhinitis,observe the changes of different metabolites after the intervention of Xiaofeng Xuanqiao Decoction,and analyze the intervention effect of Xiaofeng Xuanqiao Decoction based on metabonomics.Methods:40 rats were selected,10 were used as blank group,and the other 30 were used to establish allergic rhinitis rat model.A total of 27 rats were successfully modeled,and they were divided into the model group,western medicine group and Xiaofeng Xuanqiao Decoction group with 9 rats each.Xiaofeng Xuanqiao Decoction group was given Xiaofeng Xuanqiao Decoction,western medicine group was given montelukast sodium,blank group and model group were given 80 mg/kg normal saline.The behavioral scores of each group were evaluated on the day of successful modeling,days 1,3,and 7 after intervention,and the effects of Xiaofeng Xuanqiao Decoction on the metabolic spectrum of blood oxygen samples were analyzed.Results:Compared with the blank group,the model group,the western medicine group,and the Xiaofeng Xuanqiao Decoction group had higher behavioral scores on the day of successful modeling and on days 1,3,and 7 after intervention(P<0.05).The behavioral scores of the model group,the western medicine group,and the Xiaofeng Xuanqiao Decoction group on the day of successful modeling were not significantly different(P>0.05).Compared with the model group,the western medicine group and the Xiaofeng Xuanqiao Decoction group had lower behavioral scores on days 1,3,and 7 after intervention(P<0.05).There was no significant difference in behavioral scores between the western medicine group and the Xiaofeng Xuanqiao Decoction group on days 1,3,and 7 after intervention(P>0.05).Principal Component Analysis(PCA)showed that there were certain differences in metabolites among groups,indicating that allergic rhinitis altered the metabolism of certain substances in mouse se-rum.After intervention with Xiaofeng Xuanqiao Decoction and montelukast sodium,the altered metabolite
