摘  要:目的探究甘草黄酮对小鼠支气管肺发育不良(BPD)的保护作用及机制。方法建立BPD模型,并给予甘草黄酮灌胃治疗,于第14天收集小鼠肺组织和血清,HE染色观察小鼠肺组织形态学变化,酶联免疫吸附实验检测小鼠血清IL-18、IL-1β的表达水平,Western blot检测小鼠肺组织caspase-1、GSDMD与IL-1β的表达。结果甘草黄酮可改善BPD小鼠的肺组织损伤,降低小鼠血清促炎症因子IL-18和IL-1β的表达,降低小鼠肺组织TNF-α和IL-6蛋白表达,增加IL-10蛋白表达,降低caspase-1、GSDMD及IL-1β蛋白表达。结论甘草黄酮可通过抑制caspase-1/GSDMD/IL-1β介导的巨噬细胞焦亡改善小鼠支气管肺发育不良。Objective To investigate the protective effect and mechanism of glycyrrhiza flavone on bronchopulmonary dysplasia(BPD) in mice. Methods The BPD model was established, and the mice were given glycyrrhiza flavone by inadministration. Lung tissue and serum were collected on the 14th day, and the morphological changes of lung tissue were observed by HE staining. The levels of IL-18 and IL-1β in serum were detected by enzyme-linked immunosorbent assay. The expressions of caspase-1, GSDMD and IL-1β in lung of mice were detected by Western blot. Results Glycyrrhiza flavone can improve the lung tissue injury of BPD mice, decrease the levels of IL-18 and IL-1β in serum, decrease the expressions of TNF-α and IL-6, increase the expression of IL-10, and decrease the expressions of caspase-1, GSDMD and IL-1β protein in lung tissues of mice. Conclusion Glycyrrhiza flavone can improve bronchopulmonary dysplasia by inhibiting caspase-1/GSDMD/IL-1β mediated macrophage focal death.
