摘　　要：目的探讨不同醇类固定后涂片进行快速HE(HE)染色与D⁃Q(Diff⁃Quick)染色比较其技术指标和诊断效能的差异,评价快速HE染色在快速现场评估(ROSE)中的应用效能。方法收集160例细胞学标本进行常规涂片,分别使用甲醇和95%乙醇固定后快速HE染色、D-Q染色,在现场进行评估,比较3组ROSE脱片率、染色总耗时及诊断效能;通过镜下观察和细胞核着色平均光密度值(AOD)的计算,比较不同固定液固定效果间的差异。结果95%乙醇⁃HE ROSE组AOD值更接近经典HE,且显著高于甲醇⁃HE ROSE组,具有统计学差异(P<0.001)。3组ROSE在分级诊断符合率中C6级诊断分别为89.1%,88.0%,78.3%。结论涂片经95%乙醇固定并快速HE染色在满足ROSE时效的基础上,能够快速做出准确的诊断。Objective To investigate the differences in technical parameters and diagnostic efficacy of different alcohol⁃fixed smears subjected to rapid HE(Hematoxylin⁃Eosin,HE)staining compared with D⁃Q(Diff⁃Quick)staining and to evaluate the efficacy of rapid HE staining for use in Rapid On⁃Site Evaluation(ROSE).Methods A total of 160 cytological specimens were collected for routine smear,fixed with methanol and 95%ethanol,perform rapid HE staining and D⁃Q staining for on⁃site evaluation;the detachment rate,total staining time,and diagnostic efficacy of three groups of ROSE slides,and the differences in fixation effects were compared between different fixatives by observing under the microscope and calculating the average optical density(AOD)of cell nucleus staining.Results The AOD value of the 95%ethanol HE ROSE group was closer to classical HE and significantly higher than that of the methanol HE ROSE group,with statistical differences(P<0.001).The conformity rates of C6 grade diagnosis in the three groups of ROSE were 89.1%,88.0%,and 78.3%,respectively.Conclusions Smear fixation with 95%ethanol and rapid HE staining can make accurate diagnoses quickly while meeting ROSE time requirements.
