摘 要:采用大孔树脂柱色谱、MCI柱色谱、ODS柱色谱、半制备型高效液相色谱等方法对大血藤80%乙醇提取物进行分离纯化,采用NMR、MS等波谱方法鉴定化合物的结构,采用脂多糖(LPS)诱导的RAW264.7巨噬细胞为炎症模型评价各化合物的抗炎活性。共从中分离得到19个化合物,分别鉴定为parabaroside D(1)、(R)-2-(3,4-二羟基苯基)-2-羟乙基-O-β-D-吡喃葡萄糖苷(2)、(S)-2-(3,4-二羟基苯基)-2-羟乙基-O-β-D-吡喃葡萄糖苷(3)、原儿茶酸-3-O-β-D-葡萄糖苷(4)、对羟基苯甲酸-β-D-吡喃葡萄糖苷(5)、龙胆酸-5-O-β-D-葡萄糖苷(6)、香草酸-4-O-β-D-葡萄糖苷(7)、丁香酸葡萄糖苷(8)、尿嘧啶(9)、尿苷(10)、新绿原酸(11)、绿原酸(12)、隐绿原酸(13)、3,4-二羟基苯乙醇葡萄糖苷(14)、cuneataside A(15)、cuneataside C(16)、4-羟基-3-甲氧基苯乙酮-4-O-β-D-芹糖-(1→6)-β-D-吡喃葡萄糖苷(17)、原花青素B2(18)、baimantuoluoamide B(19)。其中,化合物1为1个新的黄烷-3-醇类成分;化合物2、3、5、6、9、19为首次从该植物中分离得到;化合物2和3为一对差向异构体。抗炎活性表明,化合物14的抗炎活性最好。Sargentodoxae Caulis was extracted with 80%ethanol and separated by macroporous resin,MCI,and ODS column chromatography and semi-preparative high performance liquid chromatography.The structures of the compounds were identified based on the NMR and MS data.A total of 19 compounds were identified as parabaroside D(1),(R)-2-(3,4-dihydroxyphenyl)-2-hydroxyethyl-O-β-D-glucopyranoside(2),(S)-2-(3,4-dihydroxyphenyl)-2-hydroxyethyl-O-β-D-glucopyranoside(3),protocatechin-3-O-β-D-glucoside(4),p-hydroxybenzoate-β-D-glucopyranoside(5),gentisic-5-O-β-D-glucopyranoside(6),vanillic acid 4-O-β-D-glucoside(7),syringic acid glucoside(8),uracil(9),uridine(10),neochlorogenic acid(11),chlorogenic acid(12),cryptochlorogenic acid(13),3,4-dihydroxyphenylethanol glucoside(14),cuneataside A(15),cuneataside C(16),4-hydroxy-3-methoxyacetophenone-4-O-β-D-apiose-(1→6)-β-D-glucopyranoside(17),proanthocyanidin B2(18),and baimantuoluoamide B(19).Compound 1 was a new flavan-3-ol,and compounds 2,3,5,6,9,and 19 were isolated from this plant for the first time.Compounds 2 and 3 were a pair of epimers.Compound 14 had the highest anti-inflammatory activity.
