摘 要:为了阐明产前瘫痪对多羔母羊血清代谢物的影响,试验将18只临产前多羔母羊分为两组,其中产前瘫痪多羔母羊(n=8)为试验组,健康妊娠多羔母羊(n=10)为对照组,两组母羊颈静脉采血后采用非靶向代谢组-液相色谱-质谱联用(LC-MS)技术分析其血清代谢物的变化,并利用metaX软件对差异代谢物进行定量和筛选。结果表明:与对照组相比,试验组血清代谢物发生明显改变,筛选出197种差异代谢物,其中105种差异代谢物在试验组中显著上调(P<0.05),包括1种乙酰乙酸、22种酰基肉碱等代谢物;92种差异代谢物在试验组中显著下调(P<0.05),包括39种溶血磷脂酰胆碱、9种溶血磷脂酰乙醇胺、4种溶血磷脂酰肌醇等代谢物。对197种差异代谢物的KEGG富集分析获得39个信号通路,并归属于七大类别,分别为细胞过程、药物开发、环境信息处理、人类疾病、新陈代谢、生物系统和遗传信息处理,主要包括酮体的合成与降解,甘油磷脂代谢,苯丙氨酸、酪氨酸和色氨酸生物合成,缬氨酸、亮氨酸和异亮氨酸降解等信号通路。说明产前瘫痪多羔母羊血清中脂类、磷脂类、酸类等代谢产物发生显著变化,并涉及多种脂质与脂类分子、氨基酸、消化系统等代谢通路。In order to clarify the effects of prenatal paralysis on serum metabolites of multiparous ewes,18 prenatal multi-lambed ewes were divided into two groups,of with the prenatal multi-lambed ewes(n=8)as the experimental group,and the healthy pregnant multi-lambed ewes(n=10)as the control group.After blood collection from jugular vein of ewes in two groups,the changes of serum metabolites were analyzed by non-targeted metabolome-liquid chromatogre-mass spectrometry(LC-MS),and the differential metabolites were quantified and screened by metaX software.The results showed that,compared with the control group,the serum metabolites in the experimental group were significantly changed,and 197 differential metabolites were selected,among which 105 differential metabolites were significantly up-regulated in the experimental group(P<0.05),including 1 acetoacetic acid and 22 acylcarnitine metabolites.A total of 92 differential metabolites were significantly down-regulated in the experimental group(P<0.05),including 39 lysophosphatidylcholine metabolites,9 lysophosphatidylethanolamine metabolites,and 4 lysophosphatidylinositol metabolites.Further KEGG enrichment analysis of 197 differential metabolites showed that 39 signaling pathways were obtained and classified into seven categories,including cellular processes,drug development,environmental information processing,human diseases,metabolism,biological systems and genetic information processing,mainly including ketone body synthesis and degradation,glycerophospholipids,phenylalanine,tyrosine and tryptophan biosynthesis,valine,leucine and isoleucine,etc.These results indicated that there were significant changes in lipid,phospholipids,acids and other metabolites in serum of prenatally paralysed multiparous ewes,which involved a variety of lipid and lipid molecules,amino acids,digestive system and other metabolic pathways.
