摘 要:为了探索猪冷冻精液最优生产流程,试验将约克夏公猪中段精液分为8组,第1组、第2组、第3组、第4组添加鸡卵黄,第5组、第6组、第7组、第8组添加鹅卵黄;第1组、第2组、第5组、第6组添加甘油后直接灌装并冷冻,第3组、第4组、第7组、第8组添加甘油后在5℃环境下平衡2 h进行灌装并冷冻;第1组、第3组、第5组、第7组解冻后不进行离心重悬,第2组、第4组、第6组、第8组解冻后在17℃条件下离心重悬。对不同组别的冻精在解冻后0小时和3小时分别进行精子活力、精子畸形率、顶体完整率等指标的检测。结果表明:在其他条件相同的情况下,加入鹅卵黄作为防冷刺激物的第5组、第6组、第7组和第8组相较于加入鸡卵黄的第1组、第2组、第3组和第4组,精子活力均显著提高(P<0.05);添加甘油后在5℃环境下平衡2 h进行灌装冷冻的第3组、第4组、第7组和第8组与直接灌装冷冻的第1组、第2组、第5组和第6组相比,在解冻后0小时的顶体完整率均差异不显著(P>0.05),在解冻后3小时测定精子活力大致呈下降趋势(第3组与第1组比,第7组与第5组比,第8组与第6组比);17℃条件下离心重悬的第2组、第4组、第6组和第8组与不进行离心重悬的第1组、第3组、第5组和第7组相比,精子活力均降低(P<0.05或P>0.05),精子畸形率显著提高(P<0.05)。使用鹅卵黄作为防冷刺激物的精液质量优于鸡卵黄;添加甘油后在5℃环境下平衡2 h进行灌装冷冻相比于直接灌装冷冻,解冻后精液质量随存放时间延长下降幅度增大;猪精液解冻后离心重悬质量明显下降。说明使用鹅卵黄,并在添加甘油后立即进行灌装冷冻和不进行离心重悬的方式可能较适合生产猪冷冻精液。In order to explore the optimal production process of frozen pig semen,the middle semen of Yorkshire boars was divided into 8 groups;groups 1,2,3 and 4 were supplemented with chicken egg yolk;groups 5,6,7 and 8 were supplemented with goose egg yolk.At the same time,groups 1,2,5 and 6 were directly filled and frozen after adding refrigerant,and groups 3,4,7,and 8 were filled and frozen by balancing at 5℃for 2 h after adding refrigerant.Groups 1,3,5 and 7 were resuspended without centrifugation after thawing;groups 2,4,6 and 8 were centrifuged and resuspended at 17℃after thawing.Sperm motility,deformity rate,and acrosome integrity rate of different groups of frozen sperm were detected at 0 h and 3 h after thawing,respectively.The results showed that under the same other condition,sperm motility in the groups 5,6,7 and 8 supplemented with goose egg yolk as anti-cold irritant substance was significantly increased compared with that in the groups 1,2,3 and 4 supplemented with chicken egg yolk(P<0.05).There was no significant difference in acrosome integrity rate at 0 h after thawing between the groups 1,2,5 and 6 that were directly filled and frozen and the groups 3,4,7,and 8 that were balanced for 2 h in a 5℃environment after adding glycerol(P>0.05),and after 3 h of thawing,the sperm motility generally showed a downward trend(group 3 vs.group 1,group 7 vs.group 5,group 8 vs.group 6).At 17℃,sperm motility in the groups 2,4,6 and 8 by centrifugation were significantly decreased compared with that in the groups 1,3,5 and 7 without centrifugation(P<0.05 or P>0.05),while sperm malformation rate was significantly increased(P<0.05).The quality of semen using goose egg yolk as anti-cold irritant substance was better than that of egg yolk.With the extension of storage time,the quality of the semen which was filled and frozen after adding glycerol and balance for 2 h in a 5℃environment decreased more significantly than that of the semen which was directly filled and frozen.After thawing,the quality of pig semen suspe
