摘 要:竹类在表观形态上的高度相似性,导致了传统形态学方法难以实现准确的竹种鉴定.为探讨DNA分子标记在竹亚科植物分类鉴定中的适用性和准确性,本研究选取了两个稳定且多态的叶绿体DNA非编码区片段(trpl32-trnL和rpl16内含子),对温州市两所高校校园园林竹种进行了序列对比和系统进化分析,并结合形态特征,鉴定所研究的样本为3属11种.从系统发育分析可知,不同属的竹种非常稳健地单独分支聚类;在遗传距离上,竹种属间遗传距离远大于种间遗传距离.此外,仅从分子标记出发,该DNA序列片段组合在属阶元达到了100%的鉴别率,但在种阶元只有27.3%的鉴别率.综上,DNA分子标记可在属阶元对竹种进行准确鉴定,再结合竹种分类图鉴,能有效提高竹种鉴定准确性和效率.Due to the high similarity in morphology of bamboo species,it is very difficult to accurately identify bamboo species by traditional morphological methods.To explore the applicability and accuracy of DNA molecular marker in the taxonomic identification of Bambusoideae,two stable and polymorphic sequences of chloroplast DNA non-coding regions(trpl32-trnL and rpl16 intron)were selected for sequence identification and phylogenetic analysis of bamboo species in two universities,Wenzhou.Coupled with morphology,these samples could be identified as 11 species belonging to 3 genera of Bambusoideae.The phylogenetic analysis showed that bamboo species of different genera were very strongly separately clustered.In terms of genetic distance,the genetic distance among different genera was far greater than that among different species.Moreover,only based on molecular marker,the combination of DNA sequences achieved a 100%identification rate at the genus level but only 27.3%at the species level.Our study shows that DNA molecular marker can accurately identify bamboo species at the genus level,and combined with the illustrated encyclopedia of bamboos,it can effectively improve the accuracy and efficiency of bamboo species classification.
