摘 要:象耳豆根结线虫(Meloidogyne enterolobii)是番茄(Solanum lycopersicum)生产中最具破坏性的病原物之一。嫁接抗性砧木可使番茄优良品种迅速获得根结线虫抗性。前期,我们鉴定了1份中抗番茄砧木品种桂砧1号。为加深对其抗性机制的理解,本研究将桂砧1号和高感品种亚拉接种象耳豆根结线虫,28 d内每隔3 d对根结占比和根系内虫数进行检测。结果表明:亚拉根结占比可高达71.7%,而桂砧1号根结占比始终保持在36.0%以下,抗、感品种根结占比在接种第12、16、20、24、28天均存在显著差异(P<0.05)。而抗、感品种根系的线虫数量在除了第20天以外的其他时间点均无显著差异。可见桂砧1号的抗性不体现在限制线虫入侵上。基于LC-MS技术对接种第5天的抗、感品种根系开展非靶向代谢组测定,以VIP>1且P<0.05为阈值,筛选出抗、感差异显著代谢物415种,其中280种上调,135种下调。差异显著代谢物种类为氨基酸及其代谢物、杂环化合物、苯及其衍生物、有机酸及其衍生物、醛酮酯类、脂肪酰类、甘油磷脂类和醇胺类等8类物质。利用KEGG数据库进行代谢通路富集分析,结果发现甘油磷脂代谢、抗坏血酸和阿糖二酸代谢、烟酸和烟酰胺代谢、甜菜素生物合成、类黄酮生物合成等代谢途径富集程度较高,以甘油磷脂代谢富集最为显著。其中磷酸胆碱、磷脂酰胆碱、溶血磷脂酰胆碱、sn-甘油-3-磷酸胆碱、三乙醇胺、二乙醇胺等物质在抗、感品种间存在显著差异。推测甘油磷脂代谢通路在一定程度上提高了桂砧1号的根结线虫抗性。本研究初步探索了番茄砧木品种桂砧1号抗象耳豆根结线虫的组织生理和代谢物基础,为后续深入研究其抗性机理提供信息参考。Meloidogyne enterolobii is one of the most devastating pathogens in tomato(Solanum lycopersicum)produc-tion.Grafting with resistant rootstocks could help superior cultivars gain root-knot nematode resistance straightly.Pre-viously,we identified a medium-resistant tomato rootstock cultivar Guizhen 1.In order to deepen the understanding to its resistance mechanism,we inoculated Guizhen 1 and susceptible cultivar Yala with M.enterolobii,and measured the proportion of root knots and nematode numbers in roots once every four days within 28 days.The root knot proportion of Yala reached 71.7%,which was kept under 36.0%in Guizhen 1.The root knot proportions between resistant and susceptible cultivars were significantly different on day 12,16,20,24 and 28 after inoculation.Whereas the differences of nematode numbers between resistant and susceptible cultivars were not significant except on day 20.Thus it can be seen that the resistance of Guizhen 1 was not reflected by limiting the invasion of nematodes.The LC-MS based non-targeted metabolome of resistant and susceptible roots on 5th day after inoculation were measured.With the threshold of VIP>1 and P<0.05,415 significantly different metabolites were gained,among which 280 were up-regulated and 135 were down-regulated.The top 8 classes of metabolites with largest number of metabolites were amino acid and its metabolites,heterocyclic compounds,benzene and its derivatives,organic acid and its derivatives,aldehyde,ketone and ester,fatty acyls,glycerol phospholipids,alcohol and amines.KEGG dataset were employed to do the metabolite pathway enrichment analysis.Glycerophospholipid metabolism,ascorbate and aldarate metabolism,nicotinate and nicotinamide metabolism,betalain biosynthesis and flavonoid biosynthesis pathways had relatively high level of enrichment,with the glycerophospholipid metabolism having the highest enrichment level.In glycerophos-pholipid metabolism pathway,there were several metabolites with significant differences between resistant and suscep-tible cultiva
