摘 要:为探明贵州烟区烟草普通花叶病毒(TMV)的差异性和多样性,本研究在贵州省贵阳市、毕节市和安顺市的4个地点收集了11个仅感染TMV的烟叶样品,并经过总RNA提取、逆转录、PCR扩增获得11个TMV样品的外壳蛋白完整序列。利用TMV外壳蛋白序列为靶标,结合系统发育分析方法,分析了贵州烟草主产烟区以及与国内主要烟区TMV之间的多样性和系统发育关系。研究结果表明:贵州烟区的TMV与国内其他省份的TMV样品之间核苷酸序列一致性为96.667%~100%,氨基酸序列一致性为98.113%~100%,存在着不同程度的亲缘关系,并存在着独特的TMV种群;贵州烟区内的TMV多样性比较明显,各主产烟区TMV之间存在着不同程度差异的遗传分化现象。本研究可为贵州不同烟区烟草普通花叶病的防治提供参考。To elucidate the heterogeneity and diversity of Tobacco Mosaic Virus(TMV)in tobacco-growing regions of Guizhou,this study collected 11 tobacco leaf samples solely infected by TMV from four locations across Guiyang,Bijie,and Anshun cities in Guizhou province.The total RNA was extracted,reverse transcribed,and PCR amplified to acquire the full coat protein sequences of the 11 TMV samples.By utilizing the TMV coat protein sequences as targets and employing phylogenetic analysis methods,we investigated the diversity and phylogenetic relationships among TMV strains in Guizhou’s primary tobacco-producing areas and those in other major domestic tobacco regions.The results indicated that the nucleotide sequence consistency between TMV in tobacco-growing regions of Guizhou and TMV samples from other provinces ranging from 96.667%to 100%,with amino acid sequence consistency ranging from 98.113%to 100%.Different levels of kinship and distinct populations of TMV were present.The diversity of TMV within Guizhou’s tobacco regions was apparent,with genetic differentiation observed to of varying degrees among TMV populations in distinct main tobacco-producing areas.This research could provide a reference for effectively preventing and controling tobacco mosaic disease in different tobacco-growing regions of Guizhou.
