摘 要:目的:研究高尿酸血症(HUA)大鼠模型在车前草水煎液干预下,机体的肾脏功能、肠道屏障完整性以及炎症因子的变化。方法:诱导HUA大鼠模型,实验结束后检测血清尿酸(UA)、肌酐(Cr)、尿素氮(BUN)、黄嘌呤氧化酶(XOD)、腺苷脱氨酶(ADA)的含量,检测肾脏尿酸转运蛋白ATP结合盒亚家族G成员2(ABCG2)、尿酸转运蛋白葡萄糖易化转运蛋白9(GLUT9)、炎症小体(NLRP3)、肿瘤坏死因子-α(TNF-α)和肠道组织紧密连接蛋白(Occludin)、紧密连接蛋白-1(Z_(O)-1)的含量。结果:车前草水煎液可降低HUA大鼠血清UA、CR、BUN水平(P<0.05),抑制XOD和ADA的活性(P<0.05),并增加ABCG2、Occludin和Zo-1的表达(P<0.05),降低NLRP3、TNF-α和GLUT9的表达(P<0.05)。结论:HUA大鼠模型出现炎症、肾和肠道功能损伤等症状,在车前草水煎液的干预下得到改善。Objective:To investigate the changes in metabolic levels and inflammatory factors in a rat model of hyperuricemia(HUA)under the intervention of Plantago asiatica decoction.Methods:The HUA rat model was induced,and after the experiment,the levels of serum uric acid(UA),creatinine(Cr),urea nitrogen(BUN),xanthine oxidase(XOD),adenosine deaminase(ADA)were detected.The levels of renal uric acid transporter ATP binding cassette subfamily G member 2(ABCG2),uric acid transporter glucose transporter 9(GLUT9),inflammasome(NLRP3),tumor necrosis factor-α(TNF-α),intestinal tissue tight junction protein(Occludin),and tight junction protein-1(Z_(O)-1)were also measured.Result:Plantago asiatica decoction can reduce the levels of serum UA,CR,and BUN in HUA rats(P<0.05),inhibit the activity of XOD and ADA(P<0.05),increase the expression of ABCG2,Occludin,and Zo-1(P<0.05),and reduce the expression of NLRP3,TNF-α,and GLUT9(P<0.05).Conclusion:The HUA rat model showed symptoms such as inflammation and renal and intestinal dysfunction,which were improved by intervention with Plantago asiatica decoction.
