基于网络药理学、分子对接和动物实验探析清热化浊方对2型糖尿病的影响及作用机制

摘　　要：为明确清热化浊方对2型糖尿病的影响和物质基础,借助网络药理学联合分子对接技术以及动物实验验证分析.发现清热化浊方治疗2型糖尿病潜在有效成分有97个,关键化合物为槲皮素,木犀草素,汉黄芩素等.候选靶点188个,关键靶点分别为TP53,AKT1,HSP90AA1等.GO富集分析显示涉及的生物功能有药物应答,氧化应激的反应,活性氧细胞代谢等.KEGG通路富集显示有脂类和动脉粥样硬化信号通路,磷脂酰肌醇信号通路等.分子对接显示有32对稳定的化合物和受体蛋白结合体.动物实验显示清热化浊方可降低DM大鼠的空腹血糖,空腹胰岛素,胰岛素抵抗指数和糖化血红蛋白,且DM大鼠肝组织中p-PI3K/PI3K,p-AKT/AKT蛋白表达显著上调(P<0.05).清热化浊方可能通过激活PI3K-AKT信号通路,改善胰岛素抵抗,从而治疗2型糖尿病,且具有作用成分多、靶点广和信号通路多样的特点.To analyze the effect,mechanism and materials of QingRe Hua Zhuo Decoction(QRHZD)in the treatment of type 2 diabetes(T2DM),basing on network pharmacology,molecular docking and rat experiment.It was found that 97 potential effective components in the treatment of type 2 diabetes by QRHZD.The inportant compounds are quercetin,luteolin,wogonin,baicalin,etc.There were 188 candidate targets and 10key target genes,including TP53,AKT1,HSP90 AA1,etc.The results of GO enrichment analysis showed that the biological functions mainly involved drug response,oxidative stress response,and cell metabolism of reactive oxygen species.KEGG pathway enrichent showed that the pathways of potential genes mainly involved lipid and atherosclerosis signaling pathways,phosphatidylinositol signaling pathways,etc.32 pairs of stable molecule compounds and receptor protein conjugates were verified by molecular docking.Animal experiments showed that Qingre Huazhuo Decoction high and low dose groups can reduce the fasting blood glucose,fasting insulin,insulin resistance index and glyco sylated hemoglobin of DM rats.Qingre Huazhuo Decoction may treat type 2 diabetes by improving insulin resis tance,and has the characteristics of many active components,rich targets and diverse signaling pathways.It may play a role mainly through PI3K-Akt signaling pathway and insulin resistance signaling pathway.
