摘 要:目的:研究熏硫前后白芷高效液相色谱法(HPLC)指纹图谱与抗偏头痛药效的相关性,筛选其熏硫加工后的差异性药效成分。方法:建立熏硫前后白芷HPLC指纹图谱,寻找其差异性化学成分。建立偏头痛大鼠模型,以血清中降钙素基因相关肽、一氧化氮、内皮素和脑组织中5-羟色胺、去甲肾上腺素的指标评价抗偏头痛药效,结合灰色关联度分析法与熵权法研究熏硫前后白芷的谱效关系,指认其差异性药效成分。结果:建立了12批白芷样品的HPLC指纹图谱,标定了13个共有峰。药效学研究结果表明,白芷熏硫前后均有抗偏头痛效果(P<0.01),其中未熏硫白芷效果更佳。谱效关联度分析结果表明,水合氧化前胡素和氧化前胡素为白芷熏硫后差异性药效成分。结论:通过谱效关系初步明确了白芷熏硫后差异性药效成分,为熏硫白芷的质量控制提供一定依据。Objective:To study the correlation between HPLC fingerprint of Angelica Dahurica Radix before and after sulfur fumigation and the anti-migraine effect,and to screen the characteristic components of the processing.Methods:The Angelica Dahurica Radix before and after sulfur fumigation HPLC fingerprints were established,the differences of chemical components between the two were searched.The migraine rat model was established.The anti-migraine efficacy was evaluated using the gene related peptide,nitric oxide,endothelin in serum and 5-hydroxytryptamine,norepinephrine in brain tissues as index,combined with the grey correlation analysis method and entropy weight method to study the spectral effect relationship,and to identify the chemical components characteristics of Angelica Dahurica Radix before and after sulfur fumigation.Results:HPLC fingerprints of 12 batches of Angelica Dahurica Radix before and after sulfur fumigation were established,and 13 common peaks were calibrated.The results of pharmacodynamic study were showed,both non-sulfurized Angelica Dahurica Radix and fumigated Angelica Dahurica Radix had anti-migraine effect(P<0.01),and non-sulfurized Angelica Dahurica Radix had better anti-migraine effect.The results of spectral correlation analysis showed that oxypeucedanin hydrate and oxypeucedanin were the different effective components of Angelica Dahurica Radix after fumigation.Conclusion:The characteristic components of Angelica Dahurica Radix after sulfur fumigation was identified by spectrum-effect relationship,which provided a certain basis for the quality control of sulful fumigated Angelica Dahurica Radix.
