摘 要:目的:探讨益气化瘀清热方通过调控SIRT1/PGC-1α线粒体生物合成通路中相关因子表达对阿霉素(ADR)诱导小鼠足细胞损伤的改善作用。方法:取分化成熟的足细胞,采用ADR诱导造模,采用siRNA构建PGC-1α转染模型,实验分为:空白组(K组),模型组(M组),益气化瘀清热方组(YQHY组),益气化瘀清热方+PGC-1α转染组(YQHY+siPGC-1α组)。采用RT-qPCR检测线粒体代谢相关基因SIRT1、PGC-1α、TFAM的表达和线粒体DNA拷贝数,线粒体呼吸链复合物Ⅰ、Ⅲ检测试剂盒检测线粒体呼吸功能。结果:与K组比较,M组和YQHY+siPGC-1α组中SIRT1、PGC-1α、TFAM mRNA表达量均显著降低(P<0.05);与M组比较,YQHY组相关mR NA表达量均显著提高(P<0.05)。与K组比较,M组和YQ H Y+s i P G C-1α组线粒体呼吸链复合物Ⅰ、Ⅲ酶活性显著降低(P<0.05);与M组比较,YQHY组线粒体呼吸链复合物Ⅰ、Ⅲ酶活性显著提高(P<0.05)。与K组比较,M组和YQHY+siPGC-1α组线粒体拷贝数表达显著降低(P<0.05);与M组比较,YQHY组表达显著升高(P<0.05)。结论:益气化瘀清热方通过调控SIRT1/PGC-1α通路,促进线粒体生物生成,减轻ADR诱导小鼠足细胞损伤。Objective:To investigate the improvement effect of Yiqi Huayu Qingre Formula on the expression of related factors in SIRT1/PGC-1αmitochondrial biosynthesis pathway in mice podocyte injury induced by adriamycin(ADR).Methods:Mature podiocytes were taken,induced by ADR,and the PGC-1αtransfection group model was established by siRNA.The experimental groups were divided into blank group(K group),model group(M group),Yiqi Huayu Qingre Formula group(YQHY group),Yiqi Huayu Qingre formula+PGC-1αtransfection group(YQHY+siPGC-1α).The expression of SIRT1,PGC-1αand TFAM genes related to mitochondrial metabolism and mitochondrial DNA copy number were detected by RT-qPCR.Mitochondrial respiratory chain complexⅠandⅢdetection kits were used to detect mitochondrial respiratory function.Results:The mRNA expressions of SIRT1,PGC-1αand TFAM in M group and YQHY+siPGC-1αgroup were significantly decreased compared with K group(P<0.05);compared with M group,the expression levels of YQHY group were significantly increased(P<0.05).Compared with K group and YQHY+siPGC-1αgroup,mitochondrial respiratory chain complexⅠandⅢenzyme activities were decreased in M group(P<0.05);compared with M group,the activity of YQHY group was increased(P<0.05).The mitochondrial copy number expression was decreased in M group and YQHY+siPGC-1αgroup compared with K group(P<0.05);compared with M group,YQHY group expressed increased(P<0.05).Conclusion:Yiqi Huayu Qingre Formula promotes mitochondrial biogenesis by regulating SIRT1/PGC-1αpathway and alleviates adriamycin-induced podiocyte injury in mice.
